Blood glucose monitor is an important part of diabetes management. With the help of blood glucose monitor results, doctors can make treatment and therapy adjustment. However, the tradition blood glucose monitoring exist the "delayed effect", which block the real-time blood glucose monitoring and the trend of blood glucose response in patients. Subcutaneous in vivo glucose biosensor, which works as an effective complement to traditional blood glucose monitoring method, can respond blood glucose fluctuation characteristics in patients precisely and avoid the risk of hypoglycemia. This method is gradually popularizing and applying in clinical.This paper takes the second generation of subcutaneous in vivo glucose sensor as the object. With the comprehensive analysis of the principle and system composition of subcutaneous glucose sensor, this paper take Pt decorated polyanilline film as the base assumption, aiming at the defects of present subcutaneous glucose biosensors. With the help of experiments, reaction parameters are improved.First, this paper analyzes the reaction principle of subcutaneous glucose biosensors and its constraints. Taking the basal layer as the research object, this paper analyzes the characteristics and methods of conducting enzyme immobilized polymer, micro/nano modified technology and comparison of a variety of conductive polymer. This paper selects the electropolymerization technology, and studies the effect of improved conductive polymer and the electrochemical doping platinum nanoparticles in glucose sensor.Through the different reaction conditions, this paper conducts several different morphologies of Pt decorated polyaniline films. Among them, polyaniline films with nanometer fiber morphology showed good electrochemical behavior and more effective electrode area.This paper studies the effect of polyaniline films with different morphologies by application testing. Test shows that, the biosensor with nano fiber morphology of polyaniline film has2-4times sensitivity than other morphologies of polyaniline prepared biosensors. Meanwhile, the sensor detect limit is0.01mmol/L, the response speed is20s. After tested in serum for3days, the sensor relative activity remains at80%. The sensor’s linear detection range is up to20mmol/L after decorated by semi-permeable membrane. The experimental results show that, nano fiber morphology Pt decorated polyaniline film has a good application prospect in glucose biosensor, and overcomes the subcutaneous glucose sensor’s low sensitivity, longer delay and linear range defects.
