Objective: This study simulates the training with the method of training thegrowing rats by jumping,and explores the effect of Electro-acupuncture on bonemetabolism of growing rats with the method of Electro-acupuncture intervention,thereby provide the theory reference for the normal bone growth and bone injury especiallyfatigue fracture prevention and treatment of adolescent athletes.Methods:40one-month old Wistar rats were randomly divided into5groups:control group, jump group, electro-acupuncture prophylaxis group,electro-acupuncture group, and spontaneous recovery group.8rats in each group.Experiments using electrical stimulation of jump training,and acupuncture interventionselection shenshu, foot sp6, YangLingQuan three chakra.Except control group,eachgroup was performed6weeks jump training of electrical stimulation. After half an hourof training on electro-acupuncture prevention group, electro-acupuncture was acted.Acupuncture therapy group start electro-acupuncture intervention after a day ofmodeling, which has3courses of treatment that includes six days(once a day except oneday rest). The study tests BMP-2and cAMP expression by microscopic, electronmicroscopic observation of tibia tissue slices and double antibody method.Results:(1) Along with the increase of time, the weight of rats showed a tendency toincrease, but the control group and prevention group weight in6weeks during theexperiment the difference was not statistically significant; The control group andjumping exercise rats weight at second weeks, the former is obviously higher than thelatter, and the difference was statistically significant (P<0.01); Prevention rats weightfrom the beginning of the third week than jumping exercise rats weight, but from the beginning of the fourth week to prevent rats weight was significantly higher in jumpingsports group, and the difference was statistically significant.(2) After6weeks jumping exercise, HE results show that jumping exercise groupcompared with the blank group lamellar bone collagen fibers arranged in distinctderangement, cortical bone density is sparse, bone cells are concentrated into paste,cortical bone as irregular, a corrugated lines calcification. Electroacupunctureprevention group than the jumping exercise rats tibia lamellar bone collagen fibersaligned slightly disordered, periosteum without visible osteolysis damage area and theproliferation of granulation tissue, the backbone of rare focal mucinous degeneration ofcortical bone density, bone cells close to the normal, uniform distribution.Electroacupuncture for treatment of rat tibial lamellar bone collagen fibers in uniformand ordered within cortical bone, bone cells are evenly distributed, the bone lacunaequantity is less, the Haversian canal without dilation, congestion, arranged in atrabecular bone cells regular ordered, uniform dispersion.(3) Tibia tissue cAMP,BMP-2expression rats in model group is significantlyhigher than those in blank control group and prevention group, and has significantdifferences (P<0.01,P<0.01);(4) Tibia tissue cAMP, BMP-2expression of rats in treatment group is significantlylower than the natural recovery group, and has significant difference (P<0.05,P<0.01).Conclusion:(1)This study with reference to domestic and foreign literature, according to HEstaining results suggest that6weeks to electrical stimulation training of jumpingexercise training to growing rats tibial stress fracture for further study of jump exerciseon bone metabolism and fatigue fracture research provides animal model.(2)Jumping exercise, on the one hand, can promote the expression of cAMP andBMP-2; and on the other hand, when cAMP and BMP-2expression can also promotebone injury occurs. Prompt that jump exercise arouse changes of bone metabolism withcAMP and BMP-2is related to the impact of the bone tissue.(3)Electro-acupuncture intervention with hypoxia.On one hand, electroacupunctureintervention can reduce jumping exercise on growing bone adverse effects on bone injury, has a certain preventive effect; on the other hand, when the bone injury,acupuncture and on bone injury treatment, and has a certain curative effect.
