1. Objective: Acute eccentric exercise can induce skeletal muscle ultrastructural damage, these micro-damage directly or indirectly induce the decline of muscle strength, symptoms of delayed onset muscle soreness and other symptoms, affect the training performance of athletes seriously. It is found that acupuncture can accelerate the recovery of skeletal muscle ultrastructure damage. This study was to observe the effects of acute eccentric exercise and acupuncture intervention on lateral force transmission of skeletal muscle, and to explore the effect of lateral force transmission in acupuncture on treatment of exercise-induced skeletal muscle damage.2. Methods: One hundred twenty male wistar rats were randomly divided into control group(C), exercise groups(E), exercise and acupuncture groups(EA), exercise and acupuncture impedance groups(EAI). Control group was allowed food and water ad libitum. In addition to control group, the other groups rats were carried out running on a treadmill down a 16°decline at 16 m·min-1 for 90 min. EA groups were used acupuncture intervention, keeping 4 minutes. EAI groups were injected with Gd Cl3 solution in the tail vein, then applied with acupuncture intervention after 30 minutes. The tissues were collected from extensor digitorum longus muscles of the rats, 1 h, 48 h, 120 h after exercise. The left EDL was used to measure the lateral force transmission. Transmission electron microscope was used to observe the right EDL ultrastructural changes, PJPs changes and ECM changes.3. Results: 3.1 The changes of total forces transmitted to the end of the EDL during series of tenotomy(αi’) and myotomy(αi) after eccentric exercise and acupuncture intervention. There is no significant difference between α3 and α3’ in control group. At 1 h, 48 h, 120 h, significant differences between α3 and α3’ in E groups and EAI groups(P<0.05、P<0.01). At 1 h, 48 h, significant differences between α3 and α3’ in EA groups, except that 120 h. 3.2 The changes of force contribution from each head(θi) after eccentric exercise and acupuncture intervention. There is no significant difference between θ2 and θ3 in control group. At 1 h, significant differences between θ2 and θ3 in E group and EAI group(P<0.01). At 48 h, EAI group got a significantly decreased(P<0.05). There is no significant difference between θ2 and θ4 in control group. At 1 h, significant differences between θ2 and θ4 in E group, EA group and EAI group. 3.3 The changes of total lateral force transmission after eccentric exercise and acupuncture intervention. Compared with control group, after an acute eccentric exercise, the rats of E groups got a substantial increase in total lateral force transmission. At 1 h, 120 h, the total lateral force transmission of EA groups were decreased comparing with E and EAI groups. At 48 h, the total lateral force transmission of EA group was higher than E group and the EAI group. EA groups were increased first and then decreased, EAI groups were continued upward trend. 3.4 The changes of skeletal muscle ultrastructure after eccentric exercise and acupuncture intervention. Compared with control group, after an acute eccentric exercise, E groups were increased in Z-lines disrupted, Z-lines destroyed, area of collagen, number of PJPs. EA groups in the Z-lines disrupted, Z-lines destroyed, number of PJPs were decreased comparing with E and EAI groups. At 1 h, 48 h, the area of collagen of EA groups were decreased comparing with E groups, but increased comparing with EAI groups. At 120 h, the area of collagen of EA group were decreased comparing with E and EAI group.4. Conclusion: 4.1 The total lateral force transmission after eccentric exercise were increased. Acupuncture can effectively regulate lateral force transmission to promote skeletal muscle injury restore. 4.2 Acupuncture can activate TRP channel of the muscle membrane, then regulate lateral force transmission of skeletal muscle, to promote muscle damage repair.
