Objective:Based on the molecular biology, through the electroacupuncture and music electroacupuncture intervention, observing and studying the chronic stress model rats cytokines IL-1β、IL-6express the change rule and its significance in hippocampal and serum, so as to further discuss the pathogenesis.Methods:Choose the healthy male SD rats,randomly divided into blank group,blank control group, model group, electroacupuncture group, music electroacupuncture group, and drug group.Using the method of chronic stress to established the depression model Through the open field experiment、 sugar water experiment and the change of the weight to observe the changes of chronic depression model rats behavior under the electroacupuncture intervene.In the hippocampal slices,using immunohistochemical methods to determine of the content of IL-1β、IL-6。 In the serum tissue, using enzyme-linked immunosorbent assay to determine of the content of IL-1β、IL-6。Results:(1) Compared with M group, the weight of K group and S group changed significantly (P<0.05);Compared with S group,the weight of K group increased slighty,but there was no statistical significance between K group and S group (P>0.05); the weight of B group. D group and Y group gained more than M group changed significantly (P<0.05) The weight of D group and B group increased less than Y group and there was significance (P>0.05)(2) Before making the models, there are no significant differences, about the crossing times,between each group(P>0.05); After making the models,the crossing times of all groups are reduced,compared with K groups, the crossing times of M group reduced significantly(P<0.05);Compared with the M group,the crossing times of B group. D group and Y group rats improved significantly (P<0.05).Compared with the B group and D group, the crossing times of Y group increased slightly, but there was no significant differences(P<0.05).(3) Before making the models, there are no significant differences, about the rearing times,between each group(P>0.05); After making the models,the rearing times of all groups are reduced,compared with K groups, the rearing times of M group reduced significantly(P<0.05);Compared with the M group,the rearing times of B group. D group and Y group rats improved significantly (P<0.05).Compared with the B group and D group, the rearing times of Y group increased slightly, but there was no significant differences(P<0.05). (4) Before making the models, there are no significant differences about the sucrose intake, between each group(P>0.05). After making the models, compared with the K group, the sucrose intake of M group decreased significantly(P<0.05); compared with the M group,the sucrose intake of B group. D group and Y group rats improved significantly (P<0.05).Compared with the B group and D group, the sucrose intake of Y group increased slightly, but there was no significant differences(P<0.05).(5) After making the models, compared with the K group,the content of IL-1βand IL-6in hippocampal and serum tissue of M group increased significantly(P<0.05); compared with the M group,the content of IL-1βand IL-6in hippocampal and serum tissue of B group. D group and Y group decreased significantly(P<0.05); compared with D group,the content of IL-1β、IL-6in hippocampal and serum tissue of Y group decreased significantly(P<0.05); compared with B group,the content of IL-1β、IL-6in hippocampal and serum tissue of Y group decreased slighty,but there was no significant difference(P>0.05).Conclusion:(1) The state of the depressing of the stress rats can be alleviated obviously by the electroacupuncture and music electroacupuncture therapy;(2) The long-time chronic stress can cause the content of IL-1βand IL-6in hippocampus and serum increased;(3) Fluovetine can alleviate the state of the depressing of the stress rats, reduce the content of IL-1βand IL-6in hippocampus and serum significantly;(4)The electroacupuncture and music electroacupuncture can alleviate the state of the depressing of the stress rats,and can reduce the content of IL-1β, IL-6in hippocampal and serum tissue,Make the immune system to reach steady state.(5)Bound state can not increase the depression state of the rats,and can not increase the content of IL-1βand IL-6in hippocampus and serum.
