| Objective:To study the effect of Guipi Decoction on depression model rat's serum 3,5,3'-triiodothyronine (T3) and the estronen receptorα(ER-α) expression character on hippocampal neurons and the mechanism is approached.Methods:①40 common female Wistar rats were selected, which having the similar weight, behavior points were divided into the control group, model group, saline group and Chinese medicine group.②Except the control group, rats in the other 3 group were established the depression model by received unpredictable stress stimulus.③No medicine was given to the control group. The model group was exerted with lavage action. The saline group was lavaged with 2ml saline. The Chinese medicine was lavaged with 0.5h earlier the stimulus.④On the 0 and 22nd day, body weight were estimated with electronic scale.⑤On the 0 and 22nd day,behavior points were estimated with open-field method.⑥The ultramicrostructure of hippocampal neurons with scanning electron microscope.⑦Serum T3 were detected with radioimmunoassay.⑧Immunohistochemistry mothod was employed to detect the ER-αexpression.⑨The data were expressed by X±S, analyzing with SPSS10.0.Result:①Compared with the control group, the body weight of model group and saline group decreased (p<0.05), and body weight of Chinese medicine group did not change much.②Compared with the control group, the behavior points of model group and saline group decreased (p<0.01), and behavior points of Chinese medicine group did not change much.③There were abundant organella in the neuron of the control and Chinese medicine groups, and the caryotheca were clear and integrity, electronic density were even in the two groups. But in the model and saline group, there were less organella, and nucleus were small, membrane were thick, perinecleus electronic density decreased.④Compared with the control group, T3 levels of the model and saline group decreased obviously (p<0.05), and that of the Chinese medicine did not change.⑤Compared with the control group, the ER-αexpression in the model and saline group decreased (p<0.01), and that in the Chinese medicine increased obviously.Conclusions:①Guipi Decoction could protect the hippocampal cells from chronic stress.②Guipi Decoction could maintain the normal excretion of T3 in the chronic stress state.③Guipi Decoction could promote the expression of ER-αin the chronic stress state.④Guipi Decoction possesses the anti-depression action, and its action mechnism is related with its maintaining the normal excretion of T3 and ER-αexpression. |
