Allergic rhinitis is an IgE-mediated symptomatic disorder of the nose due to the interaction of an allergen with IgE resulting in the release of a variety of mediators that induce the typical symptoms of rhinorrhea, sneezing, itching and nasal blockage. The prevalence of allergic rhinitis (AR) is markedly increasing worldwide as societies adopting western life styles.There’s a higher risk of asthma, conjunctivitis, chronic nasal sinusitis, adenoid hypertrophy, secretory otitis media for AR sufferers. It also induces psychological problems, such as anxiety and depression. The most introverted subjects had significantly higher scores on self reports of depression, fearfulness, and fatigue, as well as a higher prevalence of hay fever. AR markedly affects social life, sleep, school and work, and its economic impact is substantial.According to Traditional Chinese Medicine, the onset of AR requires two pathogenesis. One is the physical factors, that is deficiency in lung, spleen and kidney; the other is either the internal or external pathogenic factors,that is shown as different types of the "excess". Besides the liver plays an important role in the process of the onset and development of AR,for liver’s free coursing ensures the smooth flow of Qi,which is the foundation of internal organs’ functional activities. Heart maintains blood and vessels,which is the material basis of mind and emotion.So there is a close relationship between heart and mind. Liver is the mother organ of the heart, it stores blood and regulates the running of the blood.Therefore liver plays a role in in the development and treatment of AR,according to its regulation of lung, spleen, kidney and mind.In modern medicine, medicine is the mainly treatment of AR,which can quickly and effectively control the symptoms of AR.But it can not cure the AR sufferers, also it has a lot of side effects. Acupuncture therapy which is characterized by easy to operate, excellent effect, less adverse reaction, so it is accepted by many patients.Many ancient documents and modern papers describe the efficiency of acupuncture and moxibustion in treating allergic rhinitis. Acupuncture therapy is based on its basic pathogenesis.Conventionally it is effective according to reinforcing the lung, spleen and kidney, corresponding to dispelling wind and dampness. But it did ignore the importance of the vitality and psychological factors. In view of vitality and its interaction with AR, we designed a randomized controlled experiment of guinea pigs to investigate the efficacy and mechanism of action of acupuncture.[Objective]1. To explore whether the method of smoothing liver and regulating mind acupuncture is scientific, and the effectiveness of this treatment for AR.2. To explore the mechanism and targets of acupuncture treatment on AR, according to experimental researchMethod40guinea pigs(20males,20females) were randomly divided into five groups, Group1 (blank control group, referred to as the blank group), group2(model control group, referred to as the model group), group3(western medicine group) Group4(regular acupuncture group, referred to as the regular group), Group5(smoothing liver acupuncture group, referred to smoothing liver group), n=8,4males,4females. Group2-5copy the AR model by OVA, Observing scratching nose, sneezing, runny nose and other acts of guinea pigs that recorded symptom score for30minutes immediately after they were intranasally sensitized. AR model is successfully copied, once the guinea pig gets5scores.Interventions was carried, after the10th intranasal sensitization:blank group, model group of non-intervention; western medicine group were sprayed budesonide sprays, each nostrils spray (64ug); regular group were acupunctured bilateral Yingxiang (LI20) and Feishu (BL13); smoothing liver group were acupunctured bilateral Yingxiang (LI20), Feishu (BL13), Ganshu (BL18) and Taichong (LR3).(LI20), Feishu (BL13)and Ganshu (BL18) were without keeping the needles, the needle were kept for20minutes in LI20and LR3. Interventions for group3-5were performed once a day for15times.After the5th,10th and15th intervention, local allergens were used thrice with the guinea pigs. In group2-5saline4ml+OVA120mg (concentration3%) was used, while in blank group saline was used as a control,50ul in each nostril instillation. Immediately after the intranasal sensitization, the symptom scores within30minutes were recorded. On the day of the15th intervention, narcosis was used on the guinea pigs using10%chloral hydrate. Then blood from abdominal aorta was taken for the test of serum, IL-5, ECP and SP (ELISA). And the mucous membranes of the nasal septum were taken for nasal eosinophils (EOS) counts.ResultsWestern medicine, regular acupuncture and smoothing liver acupuncture therapy effectively decreased symptom score and inhibited the of releasing IL-5, ECP and SP, and the difference between them was not significant (p>0.05);In decreasing nasal eosinophils (EOS) cell infiltration, the smoothing liver acupuncture therapy is better than western medicine and regular acupuncture therapy(p<0.05).ConclusionSoothing liver and regulating mind acupuncture therapy may reduce EOS infiltration according to inhibiting the releasing of IL-5, ECP, SP and decrease the symptoms of AR.
