One of the most important factors for the clinical effects of acupuncture is acupuncture reinforcing and reducing manipulation. As the development of the theory of acupuncture and moxibustion, it has formed a complete theoretical system. However, various techniques from ancient medical books have too many subjective factors when using in the clinical application, limiting the acupuncture treatment in application and popularization in modern clinical. This experiment intends to figure out the possible relationship between the reinforcing and reducing manipulation of acupuncture and quantity of stimulus, like operation time, lifting-thrusting frequency and amplitude using modern scientific methods. By observing the changes of blood related indexes of blood deficiency syndrome rabbits under the different amount of stimulation, the experiment can provide the experimental basis for evaluating lifting-thrusting reinforcement and reduction scientifically.Section one:Literature SummaryThere are two literature reviews, they discuss acupuncture lifting-thrusting reinforcement and reduction and Chinese medicine syndrome of blood deficiency respectively. The first one mainly introduces the ancient theory of lifting-thrusting reinforcement and reduction, the quantity of stimulus and quantization of lifting-thrusting reinforcement and reduction, and the research profile about clinical and experimental study. The second one makes a review for the TCM syndrome of blood deficiency about its etiology and pathogenesis, treatment, modern research and animal model.Section two:Animal experimentObjective:(1) To study whether the different amount of stimulation manipulate on TCM syndrome of blood deficiency rabbits may produce different effects.(2) To study whether the operation of a small amount of stimulation may cause the better effects than the operation of a large amount of stimulation and explore the relationship between different parameters and the effect of reinforcement and reduction.(3) To evaluate objectively acupuncture manipulation of lifting-thrusting reinforcement and reduction.Methods:Sixty rabbits were randomly and evenly assigned to six groups:A group (blank group), B group (model control group), C group, D group (small amount of stimulation), E group, F group (large amount of stimulation).10rabbits per group. C to F group were called treating groups. All the group were established the blood-deficiency model by drawing blood and restricting diet besides A group. After established the blood-deficiency model, every group has the different management. Group A:not done any treatment, just fed conventionally. Group B: bound when treating groups under treatment. Group C to Group F:acupuncture both "Zusanli"(ST36) of rabbits, four different group has different treatment on lifting-thrusting time, frequency and amplitude. The amount of stimulation manipulate on each treating group from small to large is:Group C<Group D<Group E<Group F. Do the treating every other day and totally ten times. Collecting blood sample from the ear margin veins6times:before established the blood-deficiency model, after established the blood-deficiency model, after acupuncture treatment3times,7times and10times,12days after acupuncture treatment. Analyze data using SPSS17.0software.Results:(1) Before established blood-deficiency model, there are no difference in the SF and TIBC index among the five groups, P>0.05. After making the blood-deficiency model, values of SF were sharply decreased and values of TIBC grows obviously. The values of SF and TIBC in each group also has significant difference with the blank group. Therefore we can conclude the model has been established successfully.(2) After3times of acupuncture treatment, SF index:Group F is significant different to the blank group and model group, and it’s also has significant difference with the Group C and D. TIBC index:Group C, D, E, F has significant difference with the blank Group.After7times of acupuncture treatment, SF index:Group D is significant different to before treatment. TIBC index:there’s no difference between every groups.After10times of acupuncture treatment, SF index:Group C and D has significant difference with model group and before treatment, Group C has significant difference with Group F. TIBC index:there’s no significant difference among groups.12days after acupuncture treatment, SF index:Group B, C, D, E has significant difference with before treatment and Group C is significant different to blank group, Group C has significant difference with Group E and F, Group D also has significant difference to Group F. TIBC index:Group C, D, E has significant difference with before treatment, Group D and E is significant different to model group.Conclusion:(1) Using large amount and small amount of stimulus of acupuncture lifting-thrusting reinforcement and reduction method on treating blood-deficiency rabbits has different acupuncture effect. The serum ferritin of the blood-deficiency rabbits shows that the group of rabbits using the small stimulus of lifting-thrusting recover faster than the others. Therefore, we can make a conclusion that the small amount lifting-thrusting manipulation shows the acupuncture reinforcing method effect.(2) The lifting and thrusting parameters which determine the amount of stimulation of acupuncture lifting-thrusting reinforce and reduce method is correlate to the reinforcing-reducing effect. The results of this experiment show the small amount of stimulus which using small amplitude and low frequency on lifting-thrusting, and has short operation time shows the effect of reinforcing method.(3) This experiment makes precise quantification for the lifting-thrusting depth, frequency, time and amplitude of acupuncture lifting-thrusting reinforcement and reduction. This can be regarded as the preliminary discuss of acupuncture lifting-thrusting reinforcement and reduction standardization.
