The tetraploid honeysuckle called Jiufengyihao is based on the diploid honeysuckle called Damaohua as its parent. As the authorized varieties induced and bred by colchicine has advantages of high yield, good quality,saving the time of picking and strong resistance. Jiufengyihao is a new germplasm with doubled chromosome. Studies have shown that chemical components of polyploid medicinal materials will change with doubled chromosome. The medicinal materials of the tetraploid honeysuckle has sold throughout the country.But as a new germplasm, the chemical component, safety and relevant pharmacodynamics of the tetraploid honeysuckle are not clear and guaranteed..It is the reason that there is security risks existing in the clinical application. Therefore, to carry out the research work of chemistry, safety and relevant pharmacodynamics of the tetraploid honeysuckle is extremely urgent.The research object of this study was the medicinal materials of the tetraploid honeysuckle with the diploid honeysuckle which was the parent of the tetraploid honeysuckle as reference. The chromatographic fingerprints of the two germplasms were studied and on this basis whether differences of content in some main chemical compositions existed in the two germplasms were preliminarily analyzed.The content of four main chemical compositions of honeysuckle were used as evaluation index to analyze and compare the quality of the tetraploid and diploid honeysuckle. The acute toxicity study of the tetraploid honeysuckle were carried out and compared with the diploid honeysuckle to clear whether differences existed between the two germplasms.The safty of the tetraploid honeysuckle were preliminarily evaluated by means of acute toxicity test.The vitro antimicrobial study,the vitro antiviral study and antiphlogistic study in vivo of the tetraploid honeysuckle were carried out and compared with the diploid honeysuckle to clear whether differences existed between the two germplasms in these pharmacological researches. The efficacy the tetraploid honeysuckle were preliminarily evaluated by means of these pharmacological tests.The The major results were summarized as follows:1,In this paper, fourteen samples of the tetraploid honeysuckle and twelve samples of the diploid honeysuckle were collected from different regions all over the country. All the samples were the dried flower buds of honeysuckle.The stability of the two germplasms were analyzed by means of comparing the similarity of chromatographic fingerprints The analysis results showed that the chemical components of the tetraploid honeysuckle was unstable with low similarity of most regions about60percent to80percent. The chemical components of the diploid honeysuckle was stable with high similarity which was more than90percent of most regions.By observing the peak area of chromatographic fingerprints, we had found that some common peaks in the fingerprints of the two germplasms were chaged obviously.Four chemical components with good separating degree were comfirmed and the content of them were determined on the basis of research of chromatographic fingerprints.The results showed that the four chemical components of the two germplasms from different regions was different.The content of some chemical components of the tetraploid honeysuckle was higher than the diploid honeysuckle and some almost had no difference. From a chemical point of view, the tetraploid honeysuckle had good quality as medicinal materials.2. Compared with the diploid honeysuckle,we did acute toxicity test on the tetraploid honeysuckle.It had half lethal dose which were determined by preliminary test.The LD50of the tetraploid honeysuckle and the diploid honeysuckle were72.12g/kg and69.92g/kg respectively and determined by the method of Bliss.The LD50of the two germplasms were nearly412and400times as much as clinical dosage of men.It showed that the two germplasms had low acute toxicity and wide safe range.3. Compared with the diploid honeysuckle,we choosed six strains to do the vitro antimicrobial experiment on the tetraploid honeysuckle.The six strains were staphylococcus aureus, beta hemolytic streptococcus, klebsiella pneumonia, escherichia coli, pseudomonas aeruginosa and pseudomonas aeruginosa. The tetraploid honeysuckle and the diploid honeysuckle showed good bacteriostatic action and had broad antibacterial spectra.In gram-positive bacterium, the bacteriostasis intensity to beta hemolytic streptococcus and the sterilization intensity to klebsiella pneumonia of the tetraploid honeysuckle was higher than the diploid honeysuckle. In gram-negative bacterium, the sterilization intensity to escherichia coli and pseudomonas aeruginosa of the diploid honeysuckle was higher than the tetraploid honeysuckle.4. Compared with the diploid honeysuckle,we choosed the FMI influenza to do the vitro antiviral experiment.The results showed that the tetraploid honeysuckle and the diploid honeysuckle both proved inhibition to cytopathic effects of MDCK caused by FMI influenza.The inhibition concentration of the tetraploid honeysuckle were0.061mg crude drug/mL(1:4096) and0.015mg crude drug/mL(1:16384). The inhibition concentration of the diploid honeysuckle were0.488mg crude drug/mL(1:1024),0.244mg crude drug/mL(1:4096) and0.122mg crude drug/mL(1:4096). 4. Compared with the diploid honeysuckle,we choosed the models of rat foot swelling and cotton ball granuloma of rats to do the antiphlogistic experiments on the tetraploid honeysuckle to observe the acute anti-inflammatory effects and subacute anti-inflammatory effects.The results showed that the tetraploid honeysuckle had obvious anti-inflammatory action on acute and subacute inflammation.But it had no significant difference compared with the diploid honeysuckle.
