Objective: To prepare the montmorillonite-ribavinin gel ansal drops and study in vitrorelease behavior, vivo and vitro antiviral effect.Methods: Taking the compound rate as the index, through the preliminary experiment,Compare the compound rate of magnetic mixing method and ultrasonic method underneutral condition and acidic condition to determine the compound method and thecompound medium. Taking the compound rate and the antiviral effect for index, feed ratioof montmorillonite to ribavirin, compound time and compound temperature as examiningconditions in order to determine the optimum technology, prepare the montmorillonite-ribavinin compound.The montmorillonite-ribavinin gel ansal drops forming process were studied, includingmatrix preparation technology and compound added way. Use carbomer-940asmatrix,take biological adhesion, uniformity, centrifugal degree of layering as three factors,the selection of carbomer-940, glycerin, triethanolamine dosage as conditions in order toinvestigate f the optimum technology. Investigate the added way, added wet compounddirectly or drying made into powder to determine the added way.Simulate nasal environment to investigate release process, use stripping apparatus, filterpaper instead of semipermeable membrane to investigate release in vitro of the preparation.Take Respiratory Syncytial Virus as the model virus, do antiviral study in vitro underdifferent compound conditions, combining the compound rate to determine the bestoptimum technology. Set mice Respiratory Syncytial Virus model do antiviral study in vivoof montmorillonite-ribavinin gel ansal drops.Result: The optimum compound technology: ultrasonic method under acidic conditionmontmorillonite: ribavirin is1.5:1; compound temperature is70℃; compound time is3hours. The optimum technology: Carbomer-9400.25g/100ml,25%glycerol(v/v),0.3%oftriethanolamine(v/v),the compound drying24hours at80℃made into powder,then add in.The montmorillonite-ribavinin gel ansal drops have the inhibition effects on thereplication of Respiratory Syncytial Virus and obvious effect on upper respiratory tractinfection.Conclusion: The preparation of montmorillonite-ribavinin gel ansal drops has goodpreparation formability and stable character, have the inhibition effects on the replicationof Respiratory Syncytial Virus. Animal model shows that the montmorillonite-ribaviningel ansal drops obvious effect on upper respiratory tract infection. The montmorillonite-ribavinin gel ansal drops has a good application prospect.We have reached the respectedtarget.
