Objective:Dampness-constitution as a common abnormal constitution state, is considered a constitution easily happened Metabolic Syndrome. Our previous study have showed that the correlation with Metabolic Syndrome by epidemiological and experimental studies et al.The correlation of the further study Dampness-constitution with Insulin Resistance by researching conventional biochemical tests,cytokines and gene chip analysis, explore the Phlegm-dampness is the state in the development process of insulin resistance, to find an objective basis for clinical research.Method:1. Subjects were collected the morning fasting vein blood, and obtain the data of the Body Mass Index (BMI), Body Fat Percentage (F%), Waist Circumference (WC), Diastolic Blood Pressure (DBP), Systolic Blood Pressure (SBP), measure the levels of the total cholesterol (TC), triglyceride (TG), high density lipoprotein (HDL), low density lipoprotein (LDL), fasting blood glucose (FBG) and insulin (INS), calculated insulin resistance index (HOMA-IR), islet B cell function index (HOMA-B), insulin sensitivity index (ISI);2.Determination of serum leptin, resistin, adiponectin level by the Luminex-flow fluorescence techniques;3.Using gene chip technology, select the Affymetrix gene chip, extract the Phlegm-dampness crowd of peace and quality people in PBMC cells in mRNA, genome-wide detection and exploratory analysis insulin resistance related differentially expressed genes and signaling pathways;4. SPSS17.0statistical software for statistical processing. The difference was considered a statistically significant between the test.when P <=0.05.Results:1. Conventional biochemical indicators①Obesity-related indicators:the level of body mass index (BMI), waist circumference (WC), body fat percentage (F%) showed increased gradually in Balanced-constitution group, Dampness-constitution group and the MS group, there were significant differences (P<0.01or0.05).The levels of BMI, WC were significantly different compared with Balanced-constitution group (P<0.01).②Investigation on Four Items of Blood-lipid Tests:the levels of TGVTC in the Dampness-constitution group and MS group were more than Balanced-constitution group,there were significant differences (P<0.01or0.05). the levels of HDL presented a gradually decreasing trend,but there were no significant differences amongst three group.(P>0.05).③Blood Pressure:the levels of SBP, DBP presented a gradually decreasing trend,but there were no significant differences amongst three group.(P>0.05).④the comparison of insulin resistance coefficient:compared the levels of FBG\INS\HOMA-IR\ISI with Balanced-constitution group, there were significant differences (P<0.01) both in Dampness-constitution group and MS group.and there were no significant differences amongst three group in homa-b.(P>0.05).2. The levels of the cytokines Insulin resistance related leptin, resistin, adiponectin: there were no significant differences amongst three group in leptin, resistin.(P>0.05). and there were no significant differences compered the levels of ADP in Dampness-constitution group with Balanced-constitution group (P>0.05),but there were significant differences (P<0.01) with MS group.3. Genechip research result:Compared with Balanced-constitution group,there were SOCS-3/PPARA/JAK/STAT expressed increasedly,and INSIG2genes expressed decreasedly in the Dampness-constitution group.Conclusion:Phlegm-dampness has the characteristics of lipid disorders, TG, TC increased and abdominal obesity is an important feature of phlegm physical which link between obesity and insulin resistance, this stage pancreatic B cells has not been compromised, but there are ISI decline,FBG and INS raise, and then lead to the state of insulin resistance. Leptin, resistin for the time being is not yet as the markers of Phlegm-dampness link with insulin resistance, and adiponectin may be a potential marker of Phlegm-dampness and insulin resistance. Phlegm-dampness as the carrier of INSIG-2less expressed,with the related signal transduction pathway expression,and with internal factors of the formation of fat, fat metabolism disorders, link with obesity,may be the state of the progressive development ofinsulin resistance-related diseases.
