| Purpose:To observe the effect of astragalus and its effective parts on post-hemorrhagic anemia, through post-hemorrhagic anemia mice models discussing the differences and mechanism preliminarily.Material and method:132healthy SPF BALB/Conly mice, with half of female were random divided into11groups according to the hemoglobin concentration-normal group, model group, Chinese medicine control group, astragalus decocting group, ketone indican group, glucoside group, ketose group, ketoside group, polysaccharide group, flavonoids group, saponins groups,12in every group. Started to establish models after3days normal adaptive feeding. In addition to the normal group, mice in other group were bleed from orbital vein for0.5ml/kg (namely25ml/kg) on the Id,4d,7d. Teste the hemoglobin concentration on7d, the one reduced20%could be looked as post-hemorrhagic anemia model. Intragastric administrated from the2d after establishing modes, at1ml/100g weight dosage, for14d.24h after the last administration, tested RBC count peripheral blood, hemoglobin concentration, red blood cell deposited, applied ELISA on testting bone marrow stem cell factors (SCF), erythropoietin (EPO) concentration of serum and kidney tissues serous fluid, the concentration of serum specific transcription factor (GATA-1).Results:1. RBC count peripheral blood, hemoglobin concentration and red blood cells of model mice reduced significantly, which manifested that the post-hemorrhagic anemia model was successful.2. General condition:the drug treated groups could relieve the symptoms such as lacking of strength, losing weight, diet reducing due to post-hemorrhagic anemia. 3. Peripheral blood condition change:compared with model group, the treatment groups were significantly higher in hemoglobin concentrations, red blood cell count, packed-cell volume. Compared with flavonoids group, polysaccharide group, saponins group, the effect of glucoside on increasing peripheral blood red protein concentration was more significant (P<0.05).4. Compared with model group, except saponins group, serum GATA-1concentration, bone marrow SCF concentration of other treated groups were increased, and the ketose group was better than ketone indican group, glucoside group, ketose group, ketoside group, polysaccharide group, astragalus decocting group was better than ketone indican group, polysaccharide group, while the ketose group better at increasing serum GATA-1concentration than polysaccharide, astragalus decocting group was better at increasing bone marrow SCF concentration, the difference was statistically significant (P<0.05)5. Compared with model group, astragalus decocting group, ketone group, glucoside group, flavonoids group increased kidney tissues homogenate EPO concentration significantly, especially the glucoside group, the difference was statistically significant (P<0.05); Compared with model group, except for saponins, other treaed groups elevated serum EPO concentration significantly, especially the astragalus decocting group, the difference was statistically significant (P <0.05), ketose, indican compatibility is better than Single herb medicine.Conclusion:1. Astragalus and its different effective parts have the effect on normalizing post-hemorrhagic anemia in some degree.2. Astragalus and its different effective parts all can increase the peripheral blood red protein, RBC and packed-cell volume of post-hemorrhagic anemia mice, the polysaccharide+soap glucoside possibly is improves the hemorrhage anemia mouse circumference blood picture best to blend the combination effectively.3. Except saponins group, Astragalus and its different effective parts all can achieve its blood enriching functions through promoting bone marrow SCF, blood GATA-1, blood EPO, kidney tissues EPO concentration, flavonoids is the best effect to increase bone marrow SCF, blood GATA-1, polysaccharide+saponins is best effect to increase serum and kidney tissues EPO... |
