Objective:Expression of CXCL5, MMP-9proteins and mRNA were detected in gastric cancer tissuesand normal gastric tissues by immunohistochemical staining and real time quantitative PCRrespectively. So as to analyse the relationship between the expression of CXCL5and MMP-9and clinicopathological parameters and the interaction possibility between them.Material and methodsIn this study,36surgically resected gastric cancer cases were collected in the first affiliatedhosipital of Jiamusi University and Center hosipital of Jiamusi from Janury2011to May2011,each case include gastric cancer tissue and corresponding normal gastric tissues. Real-timequantitative PCR and immunohistochemical staining were used to detect expression of CXCL5and MMP-9, to detect expression of CXCL5and MMP-9protein both in the gastric cancerand in corresponding gastric tissues. The relationship between the expression of CXCL5orMMP-9and clinical pathologic parameters in the gastric cancer and the correlation betweenCXCL5and MMP-9were analysed.Result1.The results of real time quantitative PCR show that CXCL5and MMP-9wereexpressed both in the gastric cancer tissues and in normal gastric tissues, expression ofCXCL5and MMP-9was significantly higher in gastric cancer issues than them in normalgastric tissues(P<0.05).2.The results of irnrnunohistochemistry show that CXCL5mainly expressed in gastriccancer cell membrane and cytoplasm. Among the36cases,the expression of CXCL5is72.22%in gastric cancer tissues and33.33%in normal gastric tissues. expression of CXCL5ingastric cancer issues was significantly higher than that in normal gastric tissues(P<0.05). Thishave no relations with the sex, age, location of tumor and grate of differrentiation P-TNMstage while closely related to lymph node metastasise(P<0.05). The results ofirnrnunohistochemistry show that MMP-9is mainly expressed in gastric cancer cell cytoplasm,its expression among the36cases is52.8%in gastric cancer tissues while16.67%in normalgastric tissues. The expression of MMP-9have no relations with the sex, age, location of tumor (P>0.05) while closely related to grate of differrentiation, P-TNM stage andlymph node metastasise(P<0.05). The expression of MMP-9in low differentiation, P-TNMstage III+IV and have lymph node metastasis is respectively higher than that in the highdifferentiation, P-TNM stage I+II and no lymph node metastasis(P<0.05).3. Analysis of immunohistochemical staining between CXCL5and MMP-9find there wasa positive correlation between MMP-9and CXCL5proteins in gastric cancer.(r=0.2830,P=0.0945)4.There was significantly positive correlation between the mRNA and correspondingprotein expressions of CXCL5in gastric cancer tissues (P<0.05), and so is MMP-9.Conclusion1.CXCL5and MMP-9may Play an important role in the process of invasion andmetastasis of gastric cancer.2. Expression CXCL5and MMP-9protein were positively correlated,Suggesting thatthey has a certain synergy in the development of gastric cancer.3.The RNA expression and corresponding protein expression of CXCL5and MMP-9wereconsistent.4.The interaction role between CXCL5and MMP-9in gastric cancer growth,invasion,transporion protein signal pathway needs to be studied further.
