Purpose：TostudytheeffectofFuzhengRuanjianSanjiemethodontheexpressionof mRNA of hypothyroid rats myocardial myosin heavy chain alpha and beta.Material and method：Hypothyroid Wistar rats were randomly divided intomodel group, L-T4group, QiBeiFuYuanYin group. Normal group: ordinary diet,double steamed water; Model group: low iodine diet, double steamed water. L-T4group:low iodine diet,daily gavage containing0.52μg/kg levothyroxine sodiumtablets in2ml of the suspension;QiBeiFuYuanYin group:low iodine,daily gavagecontaining Chinese Herbal Medicine29.9g/kg in2ml QiBeiFuYuanYin liquid.Serumthyroid-stimulating hormone (TSH) were measured with double antibody step clipmethod ELISA method (ELISA),serum total T3(TT3), total T4(TT4) were measuredwith radioimmunoassay (RIA). Ger-Arsenite color metric method was used tomeasure median urinary iodi (MUI). Myocardial cells form were observed underLight microscopy.The expression of mRNA of myocardial myosin heavy chain alphaand beta were measured with Real-time RT-PCR.Results：1.8weeks later, compared with model group,serum TT3value of L-T4group andQiBeiFuYuanYin group increased significantly (P <0.01), no discrepancies existsamong the2groups; Serum TT4value were increased (P <0.001), no discrepanciesexists among the2groups; TSH value reduced significantly (P <0.01), thereexists discrepancies between the two groups(P <0.05), QiBeiFuYuanYin group isbetter Than L-T4group.2.8weeks later,QiBeiFuYuanYin group and L-T4group,myocardial cell morphologywere significantly improved, QiBeiFuYuanYin group is better Than L-T4group,heart rate and low voltage recovery is significantly better than the L-T4group（P＜0.001）. 3.8weeks later,The expression of α-MHC mRNA of L-T4group and QiBeiFuYuanYingroup is respectively1.77times and2.03times higher than normal group,compared with model group, both are increased, QiBeiFuYuanYin group increasedobviously（P＜0.05）;The expression of β-MHC mRNA of L-T4group andQiBeiFuYuanYin group is respectively2.61times and1.22times higher than normalgroup, compared with model group, both are reduced,but all did not drop to normallevels, QiBeiFuYuanYin group reduced obviously（P＜0.05）.Conclusion：1. Hypothyroidism cause myocardial injury, changes in the structure of themyosin.2. Fuzheng Ruanjian Sanjie method play an important role on hypothyroid ratsmyocardial injury repaired. It have important relationship with improving theα-MHC mRNA expression, reducing the β-MHC mRNA expression.
