Objective To investigate the ability of learning and memory, the changes of Aβ, PSD95and Shankl in Alzheimer’s model rats. And analyze the change and the effect of BunaoI capsule.Methods One hundred and twenty Wistar rats were randomly divided into six groups:①pseudo-operation group②model group③Naofukang group④BunaoI capsule high dose group⑤BunaoI capsule moderate dose group⑥BunaoI capsule low dose group. Twenty rats contained in each group. The rat model of AD was established by injection of D-galactose and β-amyloid rotein(Aβ1-40) in the last five groups after they were approved qualified by Morris Water Maze test. One week after the surgery, the rats underwent gavage of drugs treatment for4weeks, one time a day. The ability of learning and memory, amount of Aβ,expression level of PSD95and Shankl were observed after the treatment.Results Compared to model group, escape time and the percentage of time and swimming distance in the second and third quadrants were shortened, the percentage of time and swimming distance in the first quadrant and times of spanning the previous platform were increased in the high dose and moderate dose groups (P<0.01~P<0.05).Twenty percent marginal region percentages were reduced accompanied forty percent marginal region percentages increased in the high dose and moderate dose groups, compared to model and Naofukang groups (P<0.01-P<0.05)The proteid of Aβ in hippocampus could be reduced significantly in high dose group and moderate dose group, compared to model and Naofukang groups (P<0.05).The expression of PSD95were increased in high dose group and moderate dose group, compared to model and NimodipineNaofukang groups (P<0.01, P<0.05). The expression of Shankl were also found in moderate dose group, compared to model group and Naofukang group (P<0.01, P<0.05。Conclusion The improvement of learning and memory ability in both high dose group and moderate group, AD model established by injection of D-galactose and β-amyloid rotein (Aβ1-40), was related to Bunaol’s moderation of Aβ in hippocampus and the protied expression of PSD95and Shankl in postsynaptic dense zone.
