Objective:(1) To explore the characteristics of the local thermal effects of different moxibustion, through the observation of different moxibustion at Zusanli acupoint local temperature curve.(2)To explore the similarities and differences molecular mechanisms of the local thermal effects of different moxibustion on acupuncture points by detecting different moxibustion at Zusanli acupuncture points local Trp family gene response.Method:(1) Thirty-one healthy volunteers were treated by warming moxibustion(WM), monkshood-separed moxibustion(MM) and ginger-separed moxibustion(GM) on ST36. to observe the temperature change real-time dynamic,using of thermocouple.(2) Thirty-six Sprague-Dawley rats were randomly assigned to six groups:normal control group(NC), nomal rats with warming moxibustion treatment group (NWM), nomal rats with monkshood cake-separated moxibustion treatment group (NMM). Freund’s complete adjuvant (FCA) injection group(AA)and FCA injection with warming moxibustion treatment group (AWM) and FCA injection with monkshood cake-separated moxibustion treatment group (AMM). After administered once moxibustion treatment on ST36for15min to NWM and AWM rats, and after administered moxibustion treatment on ST36for3zhuang to NMM and AMM, the ST36tissues were isolated. We employed Trp functional classification of gene chips to detected the Trp family gene expression of acupoint ST36tissues.Result:(1).Different moxibustion have different feature of temperature.Temperature change before moxibustion at ST36was no statistical difference between3groups (p<0.05). The highest temperature in moxibustion in three groups was different significantly, GM>MM>WM (P<0.05).The temperature at the fifteenth minute after moxibustion between WM and MM was different significantly.WM>MM (P<0.05).the same to WM and GN,WM>GM(p<0.05).while in MM and GM was no statistical difference (p>0.05). (2) Under physilogical condition2DEGs(Trpv2and Trpcl) were detected, Trpv2up-regulated and Trpcl down-regulated, in monkshood cake-separated moxibustion group, and3DEGs(Trpv3,Mcolnl and Trpcl)were detected in worming moxibustion group,Trpv3up-regulated, Mcolnl and Trpcl down-regulate. Under pathological condition,2DEGs(Trpv2and Mcoln1) were detected, Trpv2up-regulated and Mcoln1down-regulated in monkshood cake-separated moxibustion group, and2DEGs(Trpv3and Trpcl)were detected in worming moxibustion group,Trpv3up-regulated, Trpcl down-regulated.(3) Trpv2on monkshood cake-separated moxibustion.and Trpv3on warming moxibustion expression levels in pathological condition than those that significantly increased trend in physiological condition.Conclusions:(1) Different moxibustion have different feature of temperature change.(2) Among Trp family gene, Trpv2, Trpv3, Mcolnl and Trpcl are shown by different moxibustion on ST36.Under the same condition, Trpv2is expressed by monkshood cake-separated moxibustion, and Trpv3is expressed by warming moxibustion. But Trpv2and Trpv3are expressed more obviously by moxibustion in pathological condition than physilogical condition.
