| 1Theoretical ResearchStress is a series of neuroendocrine reflex and changes of functions and metabolism stimulated by all kinds of factors. If the stimulation is moderate, mammals can adjust their behaviors and incretion to adapt, however, when stimulation is severe, different degrees of mental and physical dysfunction may occur. A number of experiments show that when body has been bound for a long time, functional changes caused by stress belong to the category of stagnant of liver-qi and splenic asthenia.Xiaoyaosan is a formula to cure stagnant of liver-qi and splenic asthenia. This experiment is designed to prove the effect of Xiaoyaosan on curing chronic stress.Ghrelin is a new discovered braingut petide which can stimulate growth hormone and appetite, increase fat and balance energy metabolism. Ghrelin can be found in many organs. Almost20%of ghrelin is distributed at the middle and bottom of fundic oxyntic glands and70%of ghrelin in blood circulation also comes from fundic oxyntic glands. The decrease of ghrelin can cause stagnant of liver-qi and splenic asthenia, thus both of the two diseases can be cured or improved by increasing ghrelin content.2Experimental studyThis experiment contains two parts. Experiment1is designed to show that model rats bound for21days is the model of stagnant of liver-qi/splenic asthenia by testing the change of exosyndrome, ingestion, weight, D-xylose absorption rate of small intestine.Experiment2is designed to prove the effect of xiaoyaosan on curing stagnant of liver-qi and splenic asthenia by testing the ghrelin content change in blood and bottom of fundic glands.In experiment1, randomly divide rats into3groups(24rats for each group):normal group, model group, and xiaoyaosan group. In experiment2, randomly divide rats into3groups(12rats for each group):normal group, model group and xiaoyaosan group.Bind all rats and feed xiaoyaosan group with xiaoyaosan, normal and model group with saline. Use ELISA to test amount of ghrelin in both blood and bottom of fundic glands.3Results and Conclusions3.1Experiment1Before the experiment, rats in every group are in a good condition. When bound, rats of model group and xiaoyaosan group showed strong resistance at the first time. As time passed, they adapted and put on weight slowly, while they eat and drank less than the normal group. Xiaoyaosan group performed better than the model group. During the open field test, both model group and xiaoyaosan group showed poor locomotor activity comparing with the normal group. Meanwhile, xiaoyaosan group showed better result than the model group. Xiaoyaosan played an significantly positive role in rats’weight, ingestion, open field test index and serum D-xylose content.Conclusion:Binding rats to make them stressed can cause the diseaseof stagnant of liver-qi and splenic asthenia. Through this experiment, we produced stagnant ofliver-qi/splenic asthenia model successfully.3.2Experiment2Result:After bound for21days, model group showed decrease in ghrelin content in both blood and bottom of fundic glands. As to xiaoyaosan group, ghrelin content in blood was similar with the normal group, and even more ghrelin was found in bottom of fundic glands than the amount of normal group.ConclusionXiaoyaosan can effectively cure stagnant of liver-qi and splenic asthenia by changing the ghrelin content in blood and bottom of fundic glands. |
