Influenza is the acute peogaster infectious disease which could be caused and infected rapidly and have seasonal nature. Influenza not only threatens human’s health and life but also be waste of money.In the thesis summarize, we review the knowledge and research to influenza by traditional Chinese and Western medicine first.And we also research documents about immune regulation of influenza by traditional Chinese and Western medicine second. In the exploratory chapter of the thesis, we analyse the composition, characteristics and clinical application of ChongLouKeGan dripping pills. In the thesis text, we introduce the experiment simplely as four aspects the following:Objective:We chose the mouse that infected by FM1virus strain as the model to observe the ChongLouKeGan dripping pills’effect to immunity function of the mouse model. We explore the immunity regulation and afford evidenceb for clinical application of ChongLouKeGan dripping pills.Method:1. Researching the ChongLouKeGan dripping pills how to ragulate the T-cell subpopulation of influenza virus mouse:These ICR mice are divided into six groups:that normal group, model group, ribavirin group, high dose of ChongLouKeGan dripping pills group, average dose of ChongLouKeGan dripping pills group and light ChongLouKeGan dripping pills group.Ten female mice and ten male mice in every group.They are anaesthetized by diethyl.When the mice are anaesthetized succesfully we infect them accrossing nose by4LD50virus and the dose is0.05ml.The normal group mice are injected by physiological saline with the same dose.Then these mice are intragastriced by0.3ml medicine after24h once.everyday.The normal group and model group are given by the same dose physiological saline. We treat them four days.the fourth day after injected the mice are drew blood from eye socket with no food but water in8h.We put the venous blood in anticoagulant tube for testing the quantity of T-cell subpopulation.2. Researching the ChongLouKeGan dripping pills how to affect the Inflammatory cytokine of influenza virus mouse:The method of dividing into groups,making model and treating with the same above. The mice will be put to death after be intragastriced4days.The lungs of mice are extracted asepticly,then put them into cool physiological saline for homogenating with1500rpm in centrifugal separator10minutes.At last we draw the supernatant to check the homogenating by ELISA kit.3. Researching the ChongLouKeGan dripping pills how to affect the reticuloendothelial system of influenza virus mouse:The method of dividing into groups, making model and treating with the same above. At the fourth day after treating1h, we check the phagocytose function with Carbon particle clearance method.4. Researching the ChongLouKeGan dripping pills how to affect the level of endothelin of influenza virus mouse’lung and blood plasma.The method of dividing into groups,making model and treating with the same above. The mice are drew blood1ml from eye socket after be intragastriced4days. the mice are drew blood from eye socket. We put the drew blood in the tube with10%EDTA disodium30μL and trasylol400IU,then mixed them homogeneously with3000rpm in centrifugal separator10minutes. We draw the supernatant200μl for testing. We according to the instruction of endothelin radioactive immunity-kit to test the drew bloodResult:1. The function of ChongLouKeGan dripping pills how to ragulate the T-cell subpopulation of influenza virus mouse:Compared with the normal group, the CD3+、CD4+T proportion of the model group decreases obviously(P<0.05); Compared with the model group, the CD3+、CD4+T proportion and the ratio of CD4+/CD8+of the ribavirin group increases obviously(P<0.05); the CD3+CD4+T proportion of the hight and average dose ChongLouKeGan dripping pills group increases obviously(P<0.05), the CD8+T proportion of the the average and light dose ChongLouKeGan dripping pills group decreases obviously, and the ratio of CD4+/CD8+of all the ChongLouKeGan dripping pills groups increase obviously, they all have statistic meaning(P<0.05).2. The ChongLouKeGan dripping pills how to affect the Inflammatory cytokine of influenza virus mouse:Compared with the normal group, the content of IL-10in the model group increases (P<0.05), the content of TNF-a in the model group increases evidently (P<0.001). Compared with the model group, the content of IL-4in the ribavirin and average dose ChongLouKeGan dripping pills group increases obviously(P<0.05); the content of IL-10in the hight and average dose ChongLouKeGan dripping pills group increases obviously (P<0.05); the content of IL-1β in the average and light dose ChongLouKeGan dripping pills group decreases obviously (P<0.05); the content of IL-6in the light dose ChongLouKeGan dripping pills group decreases obviously (P<0.05); the content of TNF-α in the ribavirin group decreases obviously (P<0.05); the content of TNF-α in all of the ChongLouKeGan dripping pills groups decrease evidently(P<0.001),and compared with the ribavirin group, also have statistic meaning(P<0.05).3. The ChongLouKeGan dripping pills how to affect the reticuloendothelial system of influenza virus mouse:Compared with the model group, the phagocytose index of the ribavirin group and ChongLouKeGan dripping pills groups are all elevatory and they have statistic meaning (P<0.05).4. The ChongLouKeGan dripping pills how to affect the level of endothelin of influenza virus mouse’lung and blood plasma.Compared with the normal group, the endothelin of model group are elevatory and have statistic meaning (P<0.05). Compared with the model group, the level of endothelin of the ribavirin and the average and light dose ChongLouKeGan dripping pills group decreases obviously (P<0.05).Conclusions:1. ChongLouKeGan dripping pills can regulate the immune response after the body infected with FM1by means of upregulating the CD3+、CD4+T-cell subpopulation’s percentage of the mice infected with Influnza Virus FM1and the ratios of CD4+/CD8+, meanwhile down-regulating CD8+T-cell subpopulation’s percentage.2. ChongLouKeGan dripping pills can facilitate the expressing of the anti-inflammatory Cytokines, such as IL-4、IL-10, meanwhile restrain the excessive producing of proinflammatory cytokine, such as IL-1β、IL-6、TNF-a, which balance regulate the Th type cytokines’express level of the mice infected with Influnza Virus FM1and then improve the immunologic function of the body.3. ChongLouKeGan drippig pills can activate the mononuclear phagocyte system, and enhance the reticuloendothelial system’s phagocytic function of the immuno-suppressive mice that infected with Influnza Virus FM1, furthermore make it draw near the level of the normal mice.4. ChongLouKeGan dripping pills maybe restrain the reproduction of Influnza Virus FM1, relieve the inflammatory reaction after being infected with Influnza Virus, and then decrease the expressing of ET’s mRNA and release,furthermore, alleviate the ET’s damage effect to the respiratory system.5. In conclusion, ChongLouKeGan dripping pills can ragulate the immunity inflammation influenza virus mouse with multiple target.The dripping pills can recover anti-infection of body, thus getting the antiviral function.
