OBJECTIVEFirst,through the extraorganismal experiments,to observe the cytotoxicity and the antiviral efficiency of Qiaomuqugan drop pill(QMQGDP).Second,through the Cunming mice experiments,to study the acute toxicity testing and the effectiveness of QMQGDP on influenza viral FM1 infected mice.METHODS1 The experiments on effect of QMQGDP anti-influenza virus in vitro:The cell culture technique,crystal violet staining method and MTT staining method were used to determine the inhibitory effects of Qiaomuqugan drop pill on MDCK cells activities infected by influenza virus H₁N₁,H₃N₂.Four different doses Qiaomuqugan drop pill were the treated groups and four doses Amantadine were used as the positive control groups.Besides,there are cell control group and virus control group.2 Acute toxicity test of QMQGDP in mice:The cunming mice were intragastrically administered with Qiaomuqugan drop pill at maximum concentration.There was no death of mice found and no LD₅₀ detectde after administration.Hence the maximum dosage was identified.3 Protective Effect of Qiaomuqugan drop pill on the Mice Infected with Influnza Virus FM₁:Kunming mice were inoculated intranasally with influenza virus FM₁,then treated with Qiaomuqugan drop pill,Amantadine.The experiment includes protection of Qiaomuqugan drop pill to the mice infected with influenza virus FM₁,and effect of Qiaomuqugan drop pill to the lung index and pathology of mice with influenza virus FM₁.4 The effect of QMQGDP on the lung injury and the mechanism of FM₁ infected mice: The mice were treated with Qiaomuqugan drop pill and Amantadine were used as control drugs,at the 7th day after inoculation,the mice were sacrificed,ELISA was used to measure IL-1,TNF-αin mice lung tissue homogenate and flow cytometry was used to assay T lymphocyte subsets in peripheral blood.RESULTS 1 the experiments on effect of QMQGDP anti-influenza virus in vitro:The best concentration of inhibitory multiple of Qiaomuqugan drop pill on the influenza virus H₁N₁,H₃N₂ was 0.25g/L.The treatment index(TI) on MDCK cellinfected by influenza virus H₁N₁,H₃N₂ was 14.46 and 10.32.2 Acute toxicity test of QMQGDP in mice:The maximum dosage of Qiaomuqugan drop pill is 16g/kg.3 Protective Effect of Qiaomuqugan drop pill on the Mice Infected with Influnza Virus FM₁:The life protection rate Qiaomuqugan drop pill 250mg/kg,500mg/kg groups are 55.56%,compared with model group,they are increased significantly. Compared with model group,Qiaomuqugan drop pill and Amantadine groups showed lower lung index(P<0.05) and the pathological changes were ameliorated significantly.4 The effect of QMQGDP on the lung injury and the mechanism of FM₁ infected mice: Compared with model group,lung tissue homogenate IL-1,TNF-αcontent of Qiaomuqugan drop pill and Amantadine groups were decreased significantly, (P<0.05),CD4⁺ content in peripheral blood and the ratios of CD4⁺/CD8⁺ were decreased significantly(P<0.05),CD8⁺ content was increased significantly(P<0.05).CONCLUSION1 the experiments on effect of QMQGDP anti-influenza virus in vitro: Qiaomuqugan drop pill shows strong inhibitory effects on influenza virus H₁N₁,H₃N₂ by inhibiting the proliferation of influenza virus H₁N₁,H₃N₂ and protecting the cells infected by influenza virus H₁N₁,H₃N₂.2 Acute toxicity test of QMQGDP in mice:The Qiaomuqugan drop pill is relatively safe.3 Protective Effect of Qiaomuqugan drop pill on the Mice Infected with Influnza Virus FM₁:Qiaomuqugan drop pill could significantly reduce the mortality and lung index of the mice with influenza virus FM₁ infexted and could improve pathology of the mice infected with influenza virus FM₁.So Cho- ngloukegan drop pill has a strong protective effect on the mice infected with influenza virus FM₁.4 The effect of QMQGDP on the lung injury and the mechanism of FM₁ infected mice:The Qiaomuqugan drop pill can reduce inflammatory cytokine IL-1,TNF-αand regulate the immune function of mice body,then reduces inflammatory pathology in the lung of FM1 infected mice and enhance the life protective rate.
