Abstract:Objective:To assess the treatment of xiaokanglingⅡon mouse with Ureaplasma urealyticum infection. Methods:Using standard clean grade 8~10w age of 60 healthy female mice, experimental reproductive tract swabs prior to inspection without mycoplasma infection be used as the experimental mice adapt to the environment after a week, were randomly divided into 6 groups, A group (nomal control group) 10, normal feeding; B group (model estrogen control group) 10, weekly for estrogen; C group (model group) 10, weekly for estrogen and infected with Uu; D group (western medicine treatment group) 10, weekly for estrogen, infected with Uu and to carry on two treatment courses with Azithromycin; E group (traditional Chinese medicine treatment group) 10:Weekly for estrogen, infected with Uu and to carry on two treatment courses with xiaokanglingⅡ; F group (NS control group) 10:weekly for estrogen, infected with Uu and to carry on two treatment courses with NS. B, C, D, E, FGroup cervical subcutaneous injection of estradiol benzoate per mouse 0.4 mg (0.2 mL), once a week for four weeks. B、C、D、E、F groups of mice after estrogen treatment, (1×106CCU/mL) 50μl Uu bacilli enters mice’s vagina approximately 0.5 cm with the trace pipette, direct injection, each two days time, three consecutive. Infected mice after food intake, weight, hair, activities, and external reproductive tract changes were observed and recorded weekly. Mice were inoculated after should be eluted through the vagina to train and test their Uu infection, while D group was given intraperitoneal inject tion of azithromycin, E group was given intraperitoneal injection of xiaokangling II, F group was given intraperitoneal injection of normal saline. After treating two treatment courses (one week is a treatment course), mice were killed by cervical vertebra dislocation, takes the vagina, the womb to make the tissue slice, observes its pathological change after the HE dyeing, simultaneously uses tissue culture method and immunohistochemical method to detect its Uu infection situation and use transmitssion electron microscopy to observe the ultrastructural changes. Enzymelinked immunosorbent assay (ELISA) is used to detect tumor necrosis factor-a (TNF-a) levels in serum before and after treatment. Statistical data on the use of SPSS 14.0 for windows statistical software package to carry on X2, Fish exact test and paired test for statistical analysis, p<0.05 indicates statistically significant. Results: after treatment,the positive rate of Uu s detection, traditional Chinese medicine group, western medicine group and model group, there is a significant difference (P<0.05), NS reference group and model group, there was no significant difference (P>0.05);after treatment, the TNF’s detection of mice s serum, the model control group, the NS reference group and western medicine group, there is a significant difference (P< 0.01), The model control group and the traditional Chinese medicine group compare,there is no significant difference (P> 0.05);NS reference group, western medicine and Chinese medicine group, there is a significant difference (P<0.01); the Western group and the traditional Chinese medicine group compare, there is a significant difference (P< O.01).Conclusion:Uu infection can be successfully established in mice and induce genital tract inflammation of animal model; XiaokanglingⅡhas obvious curative effect to Uu infection and can reduce the high level of TNF-a which caused by Uu infection;and xiaokanglingⅡis superior to azithromycin.
