[Objective] The purpose of this study was to study molecular biological mechanism of Shoutaiwan treating RSA by observing the effects of Shoutaiwan on the expression of TNF-α,IFN-γ,IL-4 and IL-10 in mice CD4+T cell which had a differentiation bias toward Th1 cell in vitro.[Method] Establish Th1 differentiation bias of CD4+T cell model by the activiation of ovalbumin and IL-12.These model cells were randomly divided into part A according to different concentration of drugs and part B according to different reaction times. Part A included model group,blank serum contrast group and low,middle,high concentration ShouTai Wan-containing serum groups. Part B included 0h,1h,2h,4h and 8h groups.The protein expression of TNF-α,IFN-γ,IL-4 and IL-10 were detected respectively by Western blotting.[Result] Middle and high concentration ShouTaiWan-containing serum could significantly decrease the expression level of TNF-αand IFN-γ (P<0.01).Meanwhile the high concentration ShouTaiWan-containing serum could significantly increase the expression level of IL-4 and IL-10 (P<0.01).In addition,high concentration ShouTaiWan-containing serum could significantly decrease the expression level of TNF-αand IFN-γrespectivly at 2h,4h and 8h after treating (P<0.01), the peak was 4h.High concentration ShouTaiWan-containing serum also significantly increase the expression levels of IL-4 and IL-10 respectivly at 2h and 4h (P<0.01), the peak was 4h.[Conclusion]:ShouTaiWan can suppress the expression of TNF-αand IFN-γ, promotes the expression of IL-4 and IL-10, thus promotes the differentiation of Th2, corrects the imbalance of Th1/Th2 and plays the fetus protection function.The decreasing of TNF-α, IFN-γand increasing of IL-4, IL-10 in Th cell were possiblely one of the molecular biological mechanisms for ShouTaiWan treating RSA.
