The Mechanism Study About Shoutaiwan On Adjusting Transcription Factors T-bet/GATA-3 Ratio To Reverse Th1/Th2 In RSA

Objective: To study the mRNA expressions of GATA-3,T-bet and the level of IL-2,IFN-γ,IL-4,IL-10 of blood before and after using shoutaiwan, to confirm shoutaiwan adust the ratio of T-bet/GATA-3 by upregulating expression of GATA-3 and downregulating expression of T-bet and restore Th1/Th2 cytokines balance, thus to maintain the pregnancy of habitual abortion patients.Methods: 50 RSA patients were randomly divided into two groups of A and B. Group A include 30 cases which were given shoutaiwan;group B include 20 cases which were blank control and group C include 20 healthy pregnant women. Patients in three groups were pumped peripheral venous blood two copies, one piece of blood was centrifugalized out plasma, the levels of IL-2,IFN-γ,IL-4,IL-10 of plasma were measured using ELISA method. Other piece of blood was separated PBMCs according to the density gradient centrifugation. mRNA expression of T-bet and GATA-3 of PBMCS were measured by the real-time quantitative polymerase chain reaction. Results: the level of IL-2 and INF-γwere significantly higherof patients in habitual abortion groups (group A +groupB) than those healthy pregnant women in group C, mRNA expressions of GATA-3, the level of IL-4 and IL-10 were lower to the contrary(P<0.05); The mRNA expressions of T-bet had positive correlation with the level of IL-2,IFN-γand had negative correlation with the level of IL-4,IL-10. The mRNA expressions of GATA-3 had positive correlation with the level of IL-4,IL-10 and had negative correlation with the level of IL-2,IFN-γ. The level of IL-4,IL–10 and the mRNA expressions of GATA-3 were significantly higher of group A after using shoutaiwan. The level of IL-2,IFN-γand the mRNA expressions of T-bet were ower to the contrary (P<0.05). Abortion rate was lower in group A than in group B(P<0.05).Conclusions: Shoutaiwan is a promising method for treating RSA.It can adust the ratio of T-bet/GATA-3 by upregulating the mRNA expression of GATA-3 and downregulating the mRNA expression of T-bet , thus upregulate expression of IL-4,IL-10 and downregulate expression of IL-2,IFN-γand reverse the imbalance of Th1/Th2.
