Protective Effect Of Traditional Chinese Medicine Monomer In Qidonghuoxue Oral Liquid On Acute Lung Injury Rats

Objective Previous study, we demonstrated that Qidonghuoxue Oral Liquid existence of a therapeutic effect on acute lung injury, but the specific active ingredients of drugs not yet been identified. In this study, in order to finding more effective traditional Chinese medicine monomer to the next step in-depth study, We evaluation the Qidonghuoxue Oral Liquid each monomer’s therapeutic effect in acute lung injury by animal experiments.Methods70SD rats were randomly divided into seven groups:control group,model group, solvent group, emodin group, Ligustilide groups, astragalus polysaccharide group and resveratrol group.Each group have10rats.ALI rat model prepared by Using intratracheal instillation of LPS, the solvent group by intraperitoneal injection of fat emulsion, the monomer groups by intraperitoneal injection of dose10mg/Kg in24hours before modeling, modeling, and24hours after modeling. Nothing was given in blank control group. Samples were collected48hours after modeling, then do lung tissue H&E staining, count the total number of cells and the rato of neutrophil in the bronchoalveolar lavage fluid. Detect of the total protein content and lactate dehydrogenase, the enzyme-linked immunosorbent assay in BALF and various tissues of TNF-α, IL-1β content in BALF are also assay by ELISA(enzyme-linked immunosorbent assay).Results1)in the same dose, Ligustilide can reduce the total number of cells, the ratio of neutrophils, total protein content and TNF-alpha, IL-1beta in BALF (P <0.05).2) Emodin can reduce the total number of cells and TNF-alpha and IL-1beta in BALF(P0.05), but the decrease in neutrophil ratio and total protein does not exist statistically significant (P>0.05), but it can cause significantly higher LDH level(P <0.05).3) in the same dose, APS can reduce the total protein content and TNF-alpha in BALF(P<0.05), but the reduction the ratio of the number of total cells and neutrophils was not statistically different (P>0.05).4)in the same dose, resveratrol play a a decline in the role on TNF-alpha in BALF (P<0.05), there is no significant differenceon the other indicators (P>0.05).Conclusion From the data measured in BALF,such as the total number of cells, neutrophils ratio, total protein, TNF-alpha, IL-lbeta, LDH, for acute lung injury caused by LPS, the protective effect of Ligustilide is the most completive, followed by Emodin, APS, Resveratrol...
