Objective:The thesis aims to discover the relevance between beta defensin1gene and dental caries susceptibility in the population of Han and Yugur nationality and to find out molecular genetic mechanism of caries incidence by monitoring polymorphism of three single nucleotide rs11362rs2978862and rs5743407respectively in beta-defensin1of dental caries patients and healthy control group in Gansu Yugur minority and Han nationality.Methods:The author has chosen355samples of Yugur minority and Han people in Gansu Yugur autonomous county to conduct a case-control study of which contains164samples (Han85and Yugur79samples) in case group and191samples (Han86and Yugur105samples) in control group; In addition, analyzing site genotype of three single nucleotide rs11362rs2978862and rs5743407by nucleic acid mass spectrometry technique after extrating DNA from peripheral blood; Finally, making a comparison of three locus genotype frequency, allele frequency distribution of dental caries and healthy samples in Yugur and Han people by chi-square test method and analyzing datas by SPSS16.0statistical software.Results:1)A11genotypes were in Hardy-Weinberg equilibrium range, and a linkage disequilibrium was noted between three SNPs within gene;2)Both rs11362and rs2978862SNP genotype frequencies in case and control group of Yugur were statistically significant (P<0.05). The frequency of rs11362GA genotype was lower in Yugur nationality case group (29.1%) than those in controls (48.8%) with OR=0.438(95%confidence interval [CI]=0.224-0.857), and rs2978862GA genotype was lower in Yugur nationality case group (25.3%) than those in controls (47.7%) with OR=0.396(95%confidence interval [CI]=0.201-0.783);3)No significant differences has been found in three genotype of beta-defensin1of two nationalities and Han nationality (P>0.05).Conclusion:The results suggest that genetic polymorphism in the locus of rs11362and rs2978862of gene might be associated with dental caries susceptibility in Yugur population.
