Objective:The study was on aging model rats with yang deficiency which werecaused by D-galactose and hydrogenated cortex,to explore the“ShuangguYitong” Electroacupuncture on the elderly Yang deficiency rats regulationeffects from the perspective of energy metabolism,to provide certainexperimental basis for electroacupuncture control the body’s energymetabolism to solve problems related to aging.Methods:485-month-old healthy male Sprague-Dawley(SD)rats, weight(200~230)g,were randomly divided into the normal control group(NC),the agingyang deficiency model group(Model),the electroacupuncture on “ShuangguYitong” point prescription group(EA) and the control electroacupuncturegroup (EAc),12rats in each group.The model of aged yang deficiency rats wasestablished by D-galactose forty days and hydrocortisone seven days in thelatter three groups except the normal control group.The acupoints of“Guanyuan”(GV4)、“Housanli”(ST36) and “baihui”(GV20) were used inelectroacupuncture treatment, The acupoints of “zhongji”(GV3)、“yinlingquan”(SP9) and “yintang”(GV29) were used for electroacupuncturecontrol treatment.Interconnected with a HANS-100electroacupuncture instrument,continuous wave at2HZ and1mA,15min every day,six times perweek for4weeks.After that,we observed the structure of liver mitochondrionby the transmission electron microscope,tested the activity of succinatedehydrogenase (SDH),the activity of Na+-K+-ATPase andCa2+-Mg2+-ATPase,and the content of lactic acid (LD)in the liver tissuethrough the colorimetric method,and tested the activity of lactatedehydrogenase (LDH) in the liver tissue through the microplate readermethod,explore the influence of electroacupuncture with Shuanggu Yitongpoint prescription on energy metabolism in the liver of aging yang deficiencymodel rats.Result:1.Observed the structure of liver mitochondrion by the transmission electronmicroscopeIn comparison to the normal control group,the inside and outsidemembrane of liver mitochondria was blurried,the ridge wasdeformationed,fractured or disappeard and the mitochondria was swellingedand changed,the rough endoplasmic reticulum was reduced in modelgroup.Compared with model group,electroacupuncture on “Shuanggu Yitong”point prescription group could obviously increase the number ofmitochondria,improve the morphological structure of mitochondrial,and theinside and outside membrane of liver mitochondria was clear,the ridge wascomplete,and the rough endoplasmic reticulum was no obvious changed.Themorphological structure of mitochondrial was clear and the particle oftherough endoplasmic reticulum was slightly taked off of acusector controlgroup.And the morphological structure of mitochondrial electroacupunctureon Shuanggu Yitong point prescription group was better than acusector controlgroup.2.Tested the activity of Na+-K+-ATPase and Ca2+-Mg2+-ATPase in the livertissue through the colorimetric method In comparison to the normal control group,it expressed a lower level ofNa+-K+-ATPase(P<0.05)and Ca2+-Mg2+-ATPase(P<0.01)in model group,thedifference was considered statistically significant;Compared with modelgroup,the electroacupuncture on Shuanggu Yitong point prescripttion groupcould enhance the level of Na+-K+-ATPase(P<0.05) andCa2+-Mg2+-ATPase(P<0.01),the difference was considered statisticallysignifycant;the diversity of the level of Na+-K+-ATPase andCa2+-Mg2+-ATPase between electroacupuncture on shuanggu yitong pointprescription group and acusector control group was considered statisticallysignificant(P<0.05).3.Tested the activity of succinate dehydrogenase (SDH) in the liver tissuethrough the colorimetric methodIn comparison to the normal control group,the model group expressed alower level of SDH activity,the difference was considered statisticallysignificant(P<0.01);Compared with model group,electroacupuncture onShuanggu Yitong point prescription group could enhance the level of SDHactivity,the difference was considered statistically significant(P<0.01);thediversity of the level of SDH between electroacupuncture on shuanggu yitongpoint prescription group and acusector control group was consideredstatistically significant(P<0.05).4.Tested the activity of lactate dehydrogenase (LDH) in the liver tissuethrough the microplate reader methodIn comparison to the normal control group,the model group expressed ahigher level of lactate dehydrogenase (LDH) the difference was consideredstatistically significant(P<0.01);Compared with modelgroup,electroacupuncture on Shuanggu Yitong point prescription group coulddecrease the level of the content of LDH,the difference was consideredstatistically significant(P<0.01);the diversity of the level of LDH betweenelectroacupuncture on shuanggu yitong point prescription group and acusector control group was considered statistically significant(P<0.05).5.Tested the content of lactic acid (LD) in the liver tissue through thecolorimetric methodIn comparison to the normal control group,the model group expressed ahigher level of lactic acid (LD) content,the difference was consideredstatistically significant(P<0.01);Compared with modelgroup,electroacupuncture on Shuanggu Yitong point prescription group coulddecrease the level of the content of LD,the difference was consideredstatistically significant(P<0.05);the diversity of the level of LD betweenelectroacupuncture on shuanggu yitong point prescription group and acusectorcontrol group was considered statistically significant(P<0.05).Conclusion:1.“Shuangguyitong”electroacupuncture can improve the structure of livermitochondrion in the aging yang deficiency model rats.2.The level of SDH、Na+-K+-ATPase、Ca2+-Mg2+-ATPase activity in the liverof the aging yang deficiency model rats can be enhancedby“Shuangguyitong”electroacupuncture while the level of LDH and LD arereduced,it can be concluded that the therapy of “Shuangguyitong”electroacupuncture has the function of keeping the body in the metabolicexuberant stage by improving the energy metabolism of the liver cells andstrengthening the metabolism ability of liver.3.In the aspect of improving the capability of the metabolism of theliver,“Shuangguyitong”electroacupuncture is much better than the controlelectroacupuncture group.
