Objective To research gout syndrome evidence, investigate distribution of spleen deficient. Combined genomics and metabonomics to gene expression differences of gout and metaboli-te markers, and analysis the biological characteristics. Methods To study TCM syndrome by research reference related literature mainly and explicit the syndrome manifestation. Screen the specific biomarkers related to gout by using NMR metabonomics method. We collect fecal samples because of noninvasive and more easily accepted. Based on gene microarray express-ion technology, we get gout difference genetic characteristics by comparison genetic variatio-ns between spleen deficient type gout and normal.Results We found the spleen-deficiency syndrome of gout was76.41%in total syndromes. To study the clinical features of spleen-deficiency gout patients, we found the main clinical features were loose stool, obesity, belly full, body and joints sour, etc.. In the metabolomics studies, compared with normal group, butyric acid, propionic acid, ethanol, glutamine, taurine, glucose in the gout group have obviously changed. Compared with goutnon-spleen-deficiency group, isoleucine, leucine and glycine in gout spleen-deficiency group have obviously changed. Gene microarray research show that compared with normal group, in gout spleen-deficiency group the up expression gene are509,and down expression genes are911. In gout spleen-deficiency group, the genes related to inflammatory response, lipid metabolism, drug metabolism, immune system gene expression have changed.Conclusions We found the spleen-deficiency syndrome of gout was76.41%in the total syn-dromes. The main clinical features of spleen-deficiency gout patients were loose stool, obesity, belly full, body and joints sour, etc.. Metabolomics studies have shown the obvious changed metabolites, such as butyric acid, propionic acid, ethanol, glutamine, taurine, glucose, isoleucine, leucine and glycine. According to the results of gene microarray, we get difference genetic character-ristics of gout spleen-deficiency group, and the genes related to inflammatory response, lipid metabolism, drug metabolism, immune system gene expression have changed. We found some of related changed genes in the gene microarray can be reflected in metabolites accordingly.
