Objective:To study the influence of ShuGanHeWei granules on gastrointestinal propulsion and acid reflux rat with gastro electric allorhythmia and reflux esophagitis.Methods:(1) The model rats were fed irregularly for4weeks (fasted for24h every other day), with free access to drinking water, and10mL10mol/L HCl was a dded to1L water to disturb the pH of stomach. Rat were divided into5groups:blank group, model group, high dose ShuGanHeWei granules group (10.8g/kg), low dose ShuGanHeWei granules group(5.4g/kg) and drug group (Omeprazole8.7mg/kg, Mosapride1.7mg/kg). The rats were orally administered once daily for4weeks. After treatment, Pathological changes of lower esophageal tissue were observed by using HE staining, Gastric pH, serum MTL level were determined, and dynamics of the gastric emptying and small bowel peristalsis were evaluated.(2) Reflux esophagitis model was established by pyloric ligation combined with cardiomyotomy. Rat were divided into5groups: model group, omeprazole group(8.7g/kg), ShuGanHeWei Granules groups(10.8g/kg), shamoperated group and control group. Equivoluminal drugs were adiministrated intragastic to rats,10mL/kg, once a day for4weeks. After treatment, Pathological changes and ultrastructure of LES were evaluated by using HE staining and electron microscope respectively, levels of NO, NOS, gastric pH and bilirubin, were evaluated. GAS, MTL of LES and serum gastrin were also determined.Results:(1) ShuGanHeWei Granules significantly alleviated inflammatory hyperplasia of Esophageal mucosa, lowered esophageal mucosal injured index and inflammatory responses, increased the MTL for gastric emptying and small bowel peristalsis, and improved the MTL level in serum.(P<0.01or P<0.05vs model group).(2) Ultrastructural damage including hyperplasia and inflammation of esophageal squamous epithelium, histolysis of muscle fiber in LES was improved by ShuGanHeWei Granules. In ShuGanHeWei Granules group, the levels of Gastric bilirubin, NO, NOS in LES were decreased, the levels of gastric pH, GAS, MTL in LES and serum gastrin were increased (P<0.05or P<0.01vs model group).Conclusion:By improving the gastrointestinal disorders, and inhibit acid reflux, ShuGanHeWei Granules has curative effect on rat with gastroesophageal reflux disease.
