Objective:Vertical experimental model of orthodontic tooth movement in rats,observing the movement of orthodontic force effects of topical application ofPDGF-BB, TGF-β1and their combined use on orthodontic tooth in rats, toinvestigate effects of growth factors on orthodontic tooth movement, provide thetheoretical basis for the series of Orthodontics and clinical application.Methods:From6to8weeks of age of40male Wistar rats were randomlydivided into groupA、B、C,、D,10rats in each group. Between the maxillary left firstmolar and incisor installed on a value for the orthodontic device50g, which wereinjected with the growth factor every three days in the buccal submucosal area of themolar.groupA:local injection with5ng TGF-β1and10ng PDGF-BB, groupB:localinjection with10ng PDGF-BB, groupC:local injection with5ng TGF-β1, andgroupD:local injection with physiological saline.The distance of the maxillary leftfirst molar was measured on the1,3,7,14,day.The rats were sacrificed after theexperiment lasted for fourteen days,The first molar teeth and their periodontal tissuewere observed by HE staining, periodontal tissue morphology, counter pressure sideof osteoclast number.Results:Experimental rats pressure side of osteoclast numbers in theexperiment process significantly more than the control group,And the experimentalrats teeth in the afterburning7and14day moving distance is greater than the controlgroup,PDGF-BB and TGF-β1in combined application of a synergistic effect, the pressure side osteoclast count and tooth movement distance and single factor group(P<0.05) had significant difference.Conclusion: the application of exogenous PDGF-BB and TGF-β1canincrease osteoclast generation, at the same time can significantly accelerate thedistance of tooth movement, the combination of the two, has a synergistic effect, theeffect is more obvious.
