Object:Establishment of a rat model of orthodontic tooth movement,observationof tooth movement induced alveolar bone remodeling in rat periodontal tissuespressure side, measured near the alveolar bone in periodontal membrane ofc-Fos expression in osteoclasts, through comparing c-Fos in osteoclastsexpressed within a rule, further understanding of osteoclast differentiationmechanism.Methods:Selection of95healthy Wistar rats were randomly divided into3groups:A, blank control group, B group(40g light force) and C group(100g gravityforce).Maxillary incisors is anchorage, light force and gravity forcerespectively by40g and100g force pulling the right of the maxillary first molarmesial movement. In6h,12h,24h,48h,3d,7d,14d,21d,28d produced tidduresection after periodontal, HE staining, and tartrate resistant acid phospha-tase(TRAP) staining, c-Fos staining.Results:In1,a control group of periodontal tissure pressure side occasionallyosteoclasts, c-Fos weakly positive expression.2, the experimental group of periodontal tissure pressure side of osteoclastnumbers in3d reached its peak, with time gradually decreased, and the contralgroup in21d expression had no obvious difference, the gravity groupexpression than light force group number.3, the experimental group pressure lateral periodontal membrane surr-ounding alveolar in the osteoclast immunohistochemistry showed c-Fos in 6h,12h staining pale yellow,24h,48h dyed dark brown,3d with time afterstraining gradually becomes shallow pale yellow, after7d and control grouphad no significant difference in staining;the gravity group staining than signi-ficant light force groupConclusion:In1, the same force value at different time during orthodontic toothmovement in rats with pressure lateral periodontal tissue expression of c-Fosprotein peaks before the peak of the number of osteoclast prompting c-fos inosteoclast differentiation and activation plays a very important regulatory role.In2, the same time c-Fos protein expression in the pressure side ofperiodontal tissure in orthodontic tooth movement in rats of different force ofgravity group than in the light force group, the number of osteoclast with thepressure side of periodontal tissure in orthodontic tooth movement in ratsgravity group more than in the light force group is consistent, suggesting thatc-fos in osteoclast differentiation and activation plays a very importantregulatory role.In3, c-Fos in the other cells of the periodontal membrane were alsopositive expression, tips on orthodontic tooth movement in rats model of protooncogene c-fos also may be involved in regulation of the other cells inperiodontal ligament cells to periodontal alterations.
