ObjectivesThe pathogenesis of atherosclerosis is not fully understood as yet. However, the relationship between the expression of human protection of telomeres1(hPOT1) and pathogenesis of atherosclerosis has not been described. hPOTl is the only telomere binding protein that can join at the single-stranded DNA. The other proteins are com-bined into a complex. It plays an important role in the cell division cycle, telomere length regulation, protecting the telomere ends, the maintenance of telomerase func-tion and Chromosomal atability.Methods1. Experimental patients and groups79patients were recruited to have coronary angiography. Based on the findings,49patients were taken as the study group, while the other30patients were taken as the control group. All the objects of study were evaluated the following indexes, height, weight, blood pressure, blood sugar, TC, TG, LDL-C and HDL-C.2. Measurement criteriaAll the following indexes, height, weight, blood pressure, blood sugar, TC, TG, LDL-C and HDL-C are measured in the next morning. The resting blood pressure was taken twice every other day, and compute the average systolic blood pressure. BMI=weight(kg)/hight(m)2.3. MethodsTo collect the peripheral blood of patients and healthy, determine the plasma and intracellular hPOTl level by ELISA. Total RNA was extracted by Trizol. Quantitative reverse-transcriptase polymerase chain reaction (qRT-PCR) was used to detect the relative mRNA expression. hPOTl specific primers were designed by Premer5.0, β-actin for reference.4. StatisticsStatistical analysis was performed using SPSS17.0software, measurement data was expressed as means±SD, measurement data was compared by the t-test. Linear correlation (Person-related) analysis studied the correlation between two variables. Each factor’s contribution to the atherosclerosis was determined through multivariate logistic regression. Values ofp<0.05were considered statistically significant.Results1. To measure plasma hPOTl by ELISA method, hPOT1was not detected. The re-sult has showen that hPOTl was located in cell.2. The level of intracellular hPOTl in the study group reduced significantly (P<0.05).2. The level of hPOTl mRNA in the study group reduced significantly (P<0.01).3. The level of hPOTl was negatively correlated with various risk factors. Logistic regression analysis showed that hPOT1was a protective factor for the coronary athe-rosclerotic (P<0.05).Conclusion1. Compared with the control group, the expression of hPOTl reduced significant-ly.2. The lower-expression of hPOTl was positively correlated with the occurrence and various risk factors of the coronary atherosclerotic heart disease.3. The lower-expression of hPOTl was involved in the course of the coronary atherosclerotic heart disease.
