ObjectivesThis research evaluated the effect of branch-foundation pointcombination electroacupuncture (EA) on the high-lipid feeding type insulinresistance(IR) rats,focused on exploring the expression change of IR rats’skeletal muscle GLUT4produced by branch-foundation point combinationEA and the internal mechanism involved.Methods40Wistar rats were randomly divided into2groups, normal diet groupwith16and high-lipid diet group with24, each group with female and male inaverage. Normal diet group was fed with ordinary diet, while the high-lipiddiet group was fed with high-lipid diet, and both of them were fed for8weeks.Then8rats, as well as female and male in average, selected randomlyfrom each group were conducted euglycemic-hyperinsulinemic clamptechnique (EHCT) to make sure the success of the IR model estabolishment byevaluating glucose infusion rate (GIR). The left8rats, as nomal control group,from normal diet group were sequentially fed with ordinary diet for8weeks.The left16rats from high-lipid group were randomly divided into2groups,the electroacupuncture group and the model control group, and both of themwere fed with ordinary diet for8weeks as well. At the same time the electroacupuncture group was given on the electroacupuncture treatment.After electroacupuncture theatment for6weeks, every group were conductedthe intraperitoneal insulin tolerance test (IPITT) and intraperitoneal glucosetolerance test (IPGTT) respectively to calculate the intraperitoneal insulintolerance and intraperitoneal glucose tolerance. After electroacupuncturetheatment for8weeks, measured all rats body weight (BW), fasting bloodglucose (FBG), postprandial blood glucose (PPG), combining IPITT andIPGTT, to evaluate the curative effect of EA. All rats were put to death andquickly taken the quadriceps femoris as specimens.Made use ofimmunohistochemical to detect the skeletal muscle GLUT4expression inevery group of rats, and analysed GLUT4expression level by image analysissystem.Results1. Compared the EHCT experiment conducted on normal diet group withhigh-lipid diet group showed that high-lipid diet group rats had significantlylower GIR (P <0.01); the weight of high-lipid feeding rats increasedsignificantly than ordinary feeding rats (P <0.01).2. Compared with the model control group rats, the weight, IPGT andIPIT of the electroacupuncture group reduced significantly (P <0.05, P <0.05,P <0.01); while FBG, PPG has a decreased trend, but there was nosignificant difference (P>0.05).3. Compared with normal control group, the skeletal muscle GLUT4protein expression level of the model control group decreased significantly (P<0.05);while compared with model control group, the skeletal muscleGLUT4protein expression level of the electroacupuncture group increasedsignificantly (P <0.05).Conclusions1. High-lipid diet could successfully establish insulin resistance rats model for8weeks, and was very suitable for studies of insulin resistance.2. Branch-foundation point combination electroacupuncture cansignificantly reduced IR rats’ body weight, the intraperitoneal insulintolerance and intraperitoneal glucose tolerance, IR condition could beimproved by electroacupuncture.3. Electroacupuncture could significantly contribute to the IR rats skeletalmuscle GLUT4protein expression increasing, which may be one of themechanism of remedy insulin resistance.
