Objective: To give comments on the intervention effect of auricular acupuncture on theblood glucose by adopting the techniques of Hyperinsulinemic-euglycemic clamp,it can ensure that auricular acupuncture is a way to improve rabbit’s insulin sensitivityand clarify its mechanism of the intervention effect. It provides experimental basis forauricular acupuncture to cure Type2diabetes mellitus and lays the foundation forclinical application.Methods:1. Choose12healthy rabbits from New Zealand, which were randomlydivided into non-narcotic group and the anesthesia group, and each group has6(n=6).non-narcotic group: adopting venepuncture of double auricular veins in rabbitswithout anesthesia. anesthesia group: adopting catheterization of carotid arteries andcarotid vein in rabbits under local anesthesia. To compare hyperinsulinemic-euglycemicclamp adopting venepuncture of double auricular veins in rabbits without anesthesiawith that of adopting catheterization of carotid arteries and carotid vein in rabbits underlocal anesthesia.2. Choose16healthy rabbits from New Zealand, which were randomly dividedintocontrol group and the auricular acupuncture group, and each group has8(n=8).Auricular acupuncture group: imitate the human body to find out the cymba conchae onthe both sides of rabbit acupoint, in equivalent to the human YiDian auricular point anduse electro-acupuncture treat with condensation-rarefaction wave,40minutes. Controlgroup: except not curative, it is the same as the auricular acupuncture group exceptwithout electro-acupuncture treat. Two groups use hyperinsulinemic-euglycemic clampto detect glucose infusion rate (GIR), and measure the rabbit fasting blood glucose andevaluate the plasma glucose and the steady-state plasma glucose on the rabbit InsulinSensitivity in the test period. At the same time to evaluate the rabbit’s insulin sensitivity. Results:1.Hyperinsulinemic-euglycemic adopting venepuncture of double auricularveins in rabbits without anesthesia: the rabbit plasma glucose fluctuates in the normalranges and is easy to operate with high rate of success; Hyperinsulinemic-euglycemicadopting catheterization of carotid arteries and carotid vein in rabbits under localanesthesia:the rabbit plasma glucose fluctuates largely beyond the normal ranges and iscomplex to operate with low rate of success.2.Hyperinsulinemic-euglycemic adopting venepuncture of double auricular veins inrabbits without anesthesia:the GIR coefficient of variation of the control group and theauricular acupuncture group were1.28%,0.21%. It is obvious that the auricularacupuncture group in the auricular acupuncture group was lower than that in controlgroup (P <0.05). At the same time there is no statistically significant difference aftercomparing the FBG and steady-state plasma glucose in the control group and theauricular acupuncture group (P>0.05).3. GIR of the rabbit in the auricular acupuncture group was distinctly improved, and hasstatistical significance, compared the difference with the control group was (P <0.05).4.The comparison of rabbit GIR between the control group and the auricularacupuncture group: after40minutes,50minutes, or60minutes of inputting glucose,GIR in the auricular acupuncture group increases, and the difference is statisticallysignificant (P <0.05).Conclusions:1. Hyperinsulinemic-euglycemic adopting venepuncture of double auri-cular veins in rabbits without anesthesia has certain feasibility and great stability, easyand convenient operations, low mortality rate and good repeatability. The experimentsucceeds in using the techniques of Hyperinsulinemic-euglycemic clamp on the rabbi-t.2. In the experiments blood glucose of rabbit gradually stabilizes, and the stability of theclamps in the experiment is better. While in the auricular acupuncture group, its resultshas higher stability. 3. The auricular acupuncture group has distinct difference after40minutes,50minutes,or60minutes of inputting blood glucose compared with control group. Auricularacupuncture can effectively improve the insulin sensitivity of the body and increase theutilization rate of peripheral tissue on blood glucose. It may improve insulin sensitivityby improving the liver, muscle and fat etc. peripheral tissue, regulate the body’smetabolism of blood glucose through increasing the utilization rate of around tissue, andeventually achieve the effect of lowering blood sugar.
