Hair follicles, an important appendix of mammalian skin, are composed ofseveral cell layers and have functions of maintaining homeostasis andself-thermoregulation as well as repairing skin damage. It has been proven that thereare some kinds of stem cell populations, for instance keratinocyte, mesenchymal andmelanocyte stem cells which reside in the hair follicles to serve the normal functions.Additionally, studies have shown that hair follicle-derived stem cells are easy toharvest and possess enormous proliferative capacity and the potential to differentiateinto skin tissue. Thus, we have good reason to apply hair follicle-derived stem cells toskin wound healing. Bioreactor microcarrier system is a culture technique which canexpand stem cells in quantify. The combination of bioreactor microcarrier systemwith hair follicle stem cells is ideal for expansion of cells for cell transplantation andregenerative medicine.The result showed that mesenchymal stem cells expressed high levels of CD90,CD105, CD73and CD44. They also possessed multilineage differentiation potentialfor adipogenic differentiation and osteogenic differentiation. And the expression ofCK14, CK15, CK19and Integrin β1were mostly positive in keratinocyte stem cells,but CK10and CD200were rarely positive. Bioreactor microcarrier system permittedthe cells to retain their differentiation potential and surface markers. Besides, humanhair follicle stem cells can contribute to the course of wound healing of nude mice.We harvested keratinocyte and mesenchymal stem cells. The bioreactormicrocarrier system is an efficient technique for expansion of stem cells, it permittedthe cells to retain their differentiation potential and surface markers. Human hairfollicle stem cells can contribute to the formation of new blood vessels and reduce thegranulation tissue formation.
