| The growth of hair depends on the hair follicle, and the growth of the hair follicle depends on many kinds of cells. There are different kinds of cells in the hair follicle, such as the dermal papilla cells, melanophores, the hair follicle mesenchymal stem cells and the hair follicle keratin stem cells. The hair follicle mesenchymal stem cells are one kind of adult stem cells which have the ability of selfreplication, selfrenew, proliferation, directional differentiation, and the hair follicle mesenchymal stem cells have the character of mesenchymal stem cells and the ability of be directional differentiation after the experiment of isolation, purification and identification, and this kind of cells come from the hair follicle, so we called them as the hair follicle mesenchymal stem cells. Stem cells will be relative rest state in body under a normal condition, but they will go into differentiation state if one part of body was hurted by others or effected by drugs, then the stem cells could repair the damaged cells or issues. Recently a research focus of stem cells is using stem cells to treat disease.Inducing stem cells directional differentiation by the active compounds from the Traditional Chinese Medicine becomes a research hotspot recently. It can expand pharmacological action of the Traditional Chinese Medicine, and also can study the differentiation ability of stem cells to expand their clinical using. It was found that ginsenoside and notoginsenoside could induce embryonic stem cells differentiate into vascular endothelial cells successfully through the cell signal path. Astragalus mongholicus could enhance the proliferation ability of epidermis stem cells in vitro in order to repair issues. The aqueous extraction of rhizome curculiginis induced bone marrow stem cells differentiate into neuron cells successfully. Moreover, some active compounds for Traditional Chinese Medicine, such as Radix Salviae miltiorrhiza, Scutellaria baicalensis, Quercetin also play an important role in inducing stem cells direct differentiate. These successful experiments provided theoretical evidence and experimental basis for my experiment.There were3groups in this experiment, the DMEM-F12including lOng/mL bFGF and10%FBS was positive control group, the DMEM-F12including10%FBS was negative control group, the DMEM-F12including10%FBS and gradient concertration of the active compounds of Traditional Chinese medicine were experimental groups. To test the competence of hair follicle mesenchymal stem cells of every group cultured in media including the active compounds of the Traditional Chinese Medicine after7days by MTT, and selected the active compounds of the Traditional Chinese Medicine which could improve the growth of the hair follicle mesenchymal stem cells; To test the expression of proliferating cell nuclear antigen (PCNA and Ki67) in every cell nuclear of all groups of the hair follicle mesenchymal stem cells cultured4days after drug administration by Cell Immunofluorescence, and investigate the proliferative capacity of cells from cell proliferation marker leve; to test the cell cycle percentage of every group of the hair follicle mesenchymal stem cells cultured4days by Flow cytometry, and investigate the proliferative capacity of cells from the cell cycle.The DMEM-F12including10%FBS and1μg/mL Baicailin or12.5μg/mL Ginsenosides of ginseng and leaves can improve the proliferative ability of the hair follicle mesenchymal stem cells in vitro, and so stronger than that in the DMEM-F12culture solution including10%FBS; The positive expression rates of cell nuclear PCNA and Ki67of the hair follicle mesenchymal stem cells cultured in the DMEM-F12including10%FBS and1μg/mL Baicalin or12.5μg/mL Ginseng stem-leaf saponion were similar to those of in the DMEM-F12including lOng/mL bFGF and10%FBS, the cell cycle percentages of G2and S were similar to those of the positive control group and were more than those of the negative control group; but Chenodeoxycholic Acid, Tauro ursodesoxy cholic acid and ginseng saponins Rbl have not this proliferative ability. |
