Object: In this report, human umbilical vein cell (HUVEC) is the main study object.We determined effects and mechanism of an antithrombotic traditional Chinesemedicine to anti-oxidative damage, via flow cytometry (FCM) and enzyme-linkedimmuno sorbent assay (ELISA).Methods: In this report, we divided the samples into five groups as below, the normalcontrol group, the oxidative damage model group, the low level preventive medicationgroup, the medium level preventive medication group and the high level preventivemedication group. The model and the determination of drug level: we made thehydrogen peroxide (H2O2) oxidative damage model through screening theconcentration which resulted in the oxidative damage of HUVEC, by means of cellgrowth curve. We determined the drug given level through HUVEC resuscitation,HUVEC culture and MTT. Anti-oxidative damage research in vitro: We used H2O2tomake HUVEC damaged, after given the different levels of preventive medication, andset up a normally controlled group as well as an oxidative damage model group. Fourhours later, we determined the activity of HUVEC by the means of MTT, observedthe periodic change of the cell via FCM and determined the changed level of NO,NOS by ELISA.Results: Compared with the normal control group, after4h of oxidation by0.2mmol/L H2O2, the HUVEC activity of oxidative damage model group decreasedsignificantly (P<0.01). The FCM result shows that the cell number increasedobviously during the G1phase while decreased distinctly during S phase and G2phase. And the ELISA result shows that the NO level and NOS level decreased(P<0.05). When compared with the oxidative damage model group, the HUVECactivities of the low level preventive medication group, the medium level preventivemedication group and the high level preventive medication group increased obviously (P<0.01), while the cell numbers during S phase and G2phase and the NO, NOS levelin cell increased distinctly (P<0.05).Conclusion: Xueshuan Xin-maining tablets has obviously protective effect onvascular endothelial cell. The effects can promote the HUVEC multiplication andtrigger the cell cycle progression turn to S phase G2phase, as well as increase therelease of NO and NOS.
