Copper oxide nanoparticles?CuONPs?are one of the widely used metal oxide nanoparticles.Inhaled nanoparticles can pass through the pulmonary air-blood Barrier to enter the circulatory system,leading vascular endothelial cell dysfunction and causing the development of cardiovascular diseases?CVDs?.However,the underlying molecular mechanism of CuONPs causing vascular endothelial cell death is still unclear.In the artical,our research found that:?1?Treatment of vascular endothelial cells with 20?g/ml CuONPs for 24 h can induce DNA damage and cell death;?2?CuONPs induce excessive accumulation of superoxide anion?O2-?by down-regulating NOX4,activating the oxidative stress response,thereby leading to DNA damage and cell death of vascular endothelial cells;?3?CuONPs mediate DNA damage and cell death of vascular endothelial cells by releasing copper ions and activating p38/MAPK signaling pathway.This article explores the toxic mechanism of CuONPs on vascular endothelial cells,and provides a new direction for the prevention and treatment of atherosclerosis?AS?.
