OBJECTIVE:To observe the effects of Xueshuan Xinmaining tablets onmesentery microcirculation, hemorheology and thrombosis of acute bloodstasis in rats.METHODS:Experiment One: To observe the influence of XueshuanXinmaining tablets on mesentery microcirculation in rats: Rats wererandomly divided into5groups, such as microcirculation disturbance modelgroup,Xueshuan Xinmaining (low-dose group,middle-dose group,high-dosegroup),positive medicine group(Buchang Naoxin tong capsules). Medicineswere intragastric administered to the rats consecutively for5days. Onthe5thday,After medicines administration were finished for30minutes,opened the abdomen of rats, took out the mesentery, observed mesentericdiameter of venule and arteriole, blood velocity,flow pattern,etc. When thoseparameters had stabilized, injected HMWD to induce the microcirculationdisturbance, then continued to observe and registered all of the parameters, aswell as the time of the bloodstream recovery.Experiment Two: To observe the influence on hemorheology of acuteblood stasis in rats. Rats were randomly divided into6groups, such as normalgroup,acute blood stasis model group, Xueshuan Xinmaining (low-dosagegroup,middle-dosage group,high-dosage group),positive medicine group(Buchang Naoxintong capsules). Medicines were intragastric administered tothe rats once a day. On the6thday,when medicines administration werefinished,dipped the rats into ice water, injected adrenaline hydrochloride (AD)twice to induce the acute blood stasis. On the7thday, measured the value ofrheological. Experiment Three: To observe the influence on thrombosis in vitro ofacute blood stasis in rats. The procedures of grouping, medicines irrigating,acute blood stasis model were same as experiment two. On the7thday,measured the weight and the length of the thrombosis.RESULTS:Experiment One: After the model group were injectedHMWD,the microvascular diameter attenuated, the blood velocity sloweddown distinctly, red blood cells appeared granular even choked, all that provedthe microcirculation disturbance s model was successful. At15thminute, theblood velocity and diameter of middle-dosage group and positive medicinegroup began to recover. At30thminute, both of them had recovered to thelevel before. Compared with model group, the change of those index showedobviously statistical significance(P<0.01); The blood velocity and diameter oflow-dosage group and high-dosage group began to recover at20thto25thminute. At30thminute,two groups did not recover to the levelbefore.Compared with disturbance model group, there had statisticalsignificance (P<0.05,P<0.01).Experiment Two: Compared with normal group,the whole blood viscosity,plasma viscosity in the acute blood stasis group increased badly, whichindicated the model was achieved(P<0.05,P<0.01).Compared with modelgroup,three groups of the Xueshuan Xinmaining tables could decrease thewhole viscosity (P<0.05, P<0.01).The effect on plasma viscosity was moremarked (P<0.01).Experiment Three Compared with model group, thrombus length, wetweight, dry weight of Xueshuan Xinmaining groups lessened clearly,(P<0.01).CONCLUSION:(1)Xueshuan Xinmaining tablets could improve themicrocirculation disturbance in rats. It helped blood velocity andmicrovascular diameter to recover.(2) Xueshuan Xinmaining tablets effectively ameliorate hemorheology of acute blood-stasis rats, It could reducethe blood adhesion.(3)It inhibited the thrombosis in vitro. Among of threegroups, the medium group’s effect is the best, and it is similar to BuchangNaoxintong group.
