Objective:To study the effects of rats skin fibroblasts cultured in vitro on proliferation and collagen synthesis.Methods:Culture the fibroblast of suckling rats through the technology of cell’s culturing in vitro, let active constituents of puffball act on the fibroblasts of suckling rats in different densities (37.5ug/ml、75ug/ml、150ug/ml、300ug/ml). Measuring cell’s culture by MTT, and determing the content of collagen through biotin double antibody sandwich enzyme‐linked immunosorbent method.Result:The active constituents of puffball can promote the proliferation of fibroblasts and the collagen synthesis in dose dependent way. Compared with controls,the experimental groups all show significant difference(P<0.01). With the administration dosage increased, cell growth rates and collagen secretion all tended to increase.Conclusion:Under the work of puffball in different densities,the rats fibroblasts was well activated and proliferated. |
