| (ABSTRACT) Aim Effective monomer or component was isolated and purified from Dioscorea Cirrhosa Lour. and studied on antihypertensive action to find a new medicine or lead compound for hypertensive. Obeserve its effect on blood pressure,heart rate and coronary artery flow of isolated heart. Provid the basic experiment theory for exploitate new pharmacologic action of Dioscorea Cirrhosal Lour.Methods (1) The effective chemical constituents were isolated and purified by supersound extraction, column chromatography on macroporous resin,silica gel and octadecyl silane(ODS).(2) Pursuit active site by survey pressure and heart rate with arterial carotis communis cannula of rat.(3) Detect purity quotient of isllater by chromatography techmic with TLC.(4) Deternin purity quotient by HPLC.(5) The structure was determined by spectroscopic methods,including UV, 1HNMR, 13 CNMR can MS spectrum.(6) Approach influence of active constituent in blood pressure and heart rate by dosage ca..(7) Influence of active constituent on cormary artery flow was observed with Langendorff isolated heart.Results Component A was separated and purified from n-buent of compotanol extraction.It has a single peak at 350nm by HPLC. By Hcl-Mg reaction,it can be identified as a flavanoid,which we named Dioscorea A.Although the detection of UV, 1HNMR,13CNMR and MS spectrum had be done, but the structure of component A is also in identify.By the experiment with rat ,component A can cut down blood pressure,and step down the heart rate obviously. And, it can increase coronary artery flow transient by experiment of Langendorff isolated heart.Conclusions (1) We have obtained an effective chemical component A,which is the first flavanoid extracted from Dioscorea Cirrhosa Lour.,and we named it Dioscorea A.(2) Dioscorea A cut down blood pressure .(3) Dioscorea A can step down the heart rate.(4) Dioscorea A can increase coronary artery flow transient.
