Caspase Family is the key enzyme in the process ofapoptosis,caspase-2is a highly conserved but functionally poorlydefined member of the caspase family, and it is involved in celldeath, inflammation, and differentiation.Lichen planus is a chronicor subacute inflammatory skin diseases, civatte bodies andliquefaction degeneration of basal cell is one of its typicalhistopathological features. The pathogenesis of Lichen planus is notclear. The role of caspase-2in the pathogenesis of lichen planus isworthy of further study. Objective:To explore the expression of caspase-2in lichenplanus and provide a theoretical basis to further clarify thepathogenesis of lichen planus. Methods: Formalin-fixed, paraffin-embedded tissue blocks,including28cases of lichen planus and8cases of normal skintissue, collected in the Second Hospital of Jilin Universitydermatology clinic. We apply immunohistochemical methods todetect the expression of caspase-2and use the statistical softwareto analysis the differences of the expression in the above twogroups. Results: Caspase-2was expressed in all28samples of lichenplaus, of which24cases were strongly positiveexpression.Caspase-2positive granules was observed in eachlayer of the epidermis except the cuticle, and it also be detected inthe inflammatory infiltration of superficial dermal. There are eightcases of normal skin tissue as a control group, the expression ofcaspase-2in six of them was positive, and the rest two cases werenegative.Through statistical analysis, we found that the expressionlevels of caspase-2in lichen planus is significantly higher than innormal skin tissue.Conclusion:1),Lymphocyte infiltration was found at the junctionof the dermis and epidermis.2), Caspase-2expressed in themajority of normal skin tissue, mainly located in the basal layer ofthe epidermis, it may be involved in the normal differentiation ofkeratinocytes cells.3),The expression of caspase-2is stronglypositive in every layer of the epidermis except the cuticle, and theexpression levels of caspase-2in lichen planus is significantlyhigher than in normal skin tissue. It may play an important role in theoccurrence and development of lichen planus through apoptosisand cell cycle arrest.
