| Objective:To detect the change of Yishenjiangzhuo granules on bone gla protein (BGP), Ca、 P、Interleukin-6(IL-6), Interleukin-10(IL-10)、B lymphocytes rate(CD19+), B lymphocytes activation rate(CD19+CD69+), apoptotic rate(AV) and regulatory T cells(Treg) in patients with chronic kidney disease(CKD) stage3-4before and after treatment. To explore the influence of Yishenjiangzhuo granule on bone metabolic indicators and immune indices in patients of CKD stage3-4.Methods:Fifty-three patients with chronic renal disease were randomly divided into YSJZ group and control group. Serum level of BGP was determined by ELISE assay. The level of peripheral blood B lymphocytes activation rate(CD19+CD69+), apoptotic rate(AV), Treg (CD4+CD25+CD127lo) was detected by three-color fluorescent labeling flow cytometry. IL-6, IL-10levels were detected by CBA flow analysis system;blood contents of calcium (Ca),phosphate (P),blood urea nitrogen (BUN),creatinine (Scr) by automatic biochemical analyser.Results:①Compared with normal group, levels of BGP in both groups before treatment were higher (P<0.05); Compared with before treatment, BGP level showed no significant change after treatment (P>0.05).②After treatment, Ca increased in the two groups (P<0.05), but the treatment group had no significant difference between the control group (P>0.05); The levels of P and calcium-phosphorus product had no significant change After treatment,(P>0.05).③After treatment the level of CD19+CD69+in both groups showed no significant change(P>0.05); The level of CD19+AV in both groups were higher than normal group, whereas the CD19+AV level was lower、CD19+level was higher after treatment in both group.Significant changes of CD19+AV and CD19+in the YSJZ group compared with control group (P<0.05).④L-6level in both YSJZ and control group treatment before treatment were higher than normal group (P<0.05); Compared with the IL-6、IL-10levels before treatment, no significant difference were found after treatment in both group.⑤After treatment,the level of Treg showed no significant change in the both groups(P>0.05), but Treg level in both YSJZ group and control group were lower Compared with normal group (P <0.05).⑥After treatment Scr decreased in both groups (P<0.01),while the YSJZ group decreased more significantly (P<0.05).While CCr increased in both groups (P<0.01), while the YSJZ group increased more significantly (P<0.05).⑦orrelation analysis:negative significant correlation were observed between Scr and Ccr, BUN and Ccr, CD19+AV and Ca×P (P<0.05); positive significant correlation were observed between BUN and Ca×P (P<0.05)。Conclusions:1. Levels of BGP were increased, and bone metabolic indicators disordered in patients with chronic kidney disease(CKD) stage3-4.2. Levels of IL-6、CD19+AV increased in CKD stage3-4patients, indicated their immune system was actived.3. The activation of immune system in patients with CKD Stage3-4involved in inducing osteoclasts activation.4. Yishen Jiangzhuo Granule could improve kidney function and immune state in CKD patients,but could not improve the BGP level. |
