Objective:Esophageal carcinoma is one of the high incidence carcinomas in malignant digestivetumors in our country.Patients with esophageal carcinoma were died from metastasis ofesophageal carcinoma.To investigate the expression of amplified in breast cancer1(AIB1),matrix metalloproteinases-2(MMP-2) and Ki67in esophageal normal tissues、adjacent tissues and esophageal squamous cell carcinoma and analysis their function incarcinogensis,development,invasion and metastasis of esophageal squamous cellcarcinoma.To explore relationships betwee them in order to find out useful ways forcarcinogensis,prognosis, gene targeting therapy ofesophageal squamous cell carcinoma.Methods:41cases of esophageal carcinoma were collected from Tengzhou People Hospital in2002from2007.1to2010.12.And select the normal medical20cases of normalesophageal squamous epithelial tissue as the control group.The expression of AIB1,MMP-2protein, Ki67were detected by immunohistochemical ldpe-g-nvp.To investigatethe relationship between the positive expression of AIB1, MMP-2, Ki67and clinicalpathology by compared the expression of AIB1, MMP-2protein, Ki67in esophagealnormal tissues、adjacent tissues and esophageal squamous cell carcinoma.Results:(1) The expression of AIB1in esophageal squamous cell carcinoma (48.3%,29/60)was significantly higher than the adjacent tissues (10%,6/60) and normal tissue (5%,1/20)(P<0.05). There was only one case of low expression of AIB1in20cases of normaltissue.There was6cases of low expression of AIB1in60cases of esophageal canceradjacent tissues. There was not significant difference of the expression of AIB1in the adjacent tissues and normal tissue (P>0.05). There were20cases of positiveover-expression of AIB1in60cases of esophageal carcinoma.The expression of AIB1inesophageal squamous cell carcinoma Ⅲ~Ⅳ midterm was significantly higher thantheⅠ~Ⅱ period (P<0.05). The positive expression of AIB1in the low degree ofdifferentiation was higher than the the high degree (P<0.05). And The positive expressionof AIB1in lymph node metastasis was significantly higher (P<0.05). There was notsignificant difference of the positive expression of AIB1in gender,age and pathologicaltype (P<0.05).(2) The expression of MMP-2in esophageal squamous cell carcinoma (70.0%,42/60)was significantly higher than the adjacent tissues (16%,10/60) and normal tissue (5%,1/20)(P<0.05). The content of MMP-2in adjacent noncancerous tissues was higher thannormal tissue, but the difference was not significantly (P>0.05). The expression of MMP-2in esophageal squamous cell carcinoma Ⅲ~Ⅳ midterm was significantly higher thantheⅠ~Ⅱ period (P<0.05). The positive expression of MMP-2in the low degree ofdifferentiation was higher than the the high degree (P<0.05). And The positive expressionof MMP-2in lymph node metastasis was significantly higher than those without lymphnode metastasis (P<0.05). There was not significant difference of the positive expressionof MMP-2in gender and age(P>0.05).(3) The expression of Ki67in esophageal squamous cell carcinoma (65.0%,39/60)was significantly higher than the adjacent tissues (18.3%,11/60) and normal tissue (15.0%,3/20)(P<0.05). The content of Ki67in adjacent noncancerous tissues was higher thannormal tissue, but the difference was not significantly (P>0.05).The expression of Ki67inesophageal squamous cell carcinoma Ⅲ~Ⅳ midterm was significantly higher thantheⅠ~Ⅱ period (P<0.05). The positive expression of Ki67in the low degree ofdifferentiation was higher than the the high degree (P<0.05). And The positive expressionof Ki67in lymph node metastasis was significantly higher than those without lymph nodemetastasis (P<0.05). There was not significant difference of the positive expression ofKi67in gender and age(P>0.05).(4) There was a correlation between the expression of AIB1and MMP-2.42cases ofthe expression of MMP-2were positive in60cases of esophageal squamous cellcarcinoma, in which27cases of the expression of AIB1were positive, especially25casesof overexpression. Through the disaggregated data, there was a positive correlationbetween the expression of AIB1and MMP-2(P<0.01).But only12cases in31cases ofnegative AIB1expression had positive expression of Ki67. Through the analysis,there was a positive correlation between the expression of AIB1and MMP-2(P<0.01, r=0.438).(5) There was a correlation between the expression of AIB1and Ki67. There was thepositive expression of Ki67in28cases of simultaneous expression of AIB1of39cases inesophageal squamous cell carcinoma tissue samples, and25cases of AIB1wereoverexpression. Through the disaggregated data, there was a positive correlation betweenthe expression of AIB1and Ki67(P<0.01).28cases of Ki67expression were positive in29cases of AIB1positive expression.But only11cases in31cases of negative AIB1expression had positive expression of Ki67. Through the analysis,there was a positivecorrelation between the expression of AIB1and Ki67(P<0.01).Conclusions:(1) The expression of AIB1in esophageal squamous cell carcinoma is significantlyupregulated.The expression of AIB1was related with the clinic staging, clinical stages andthe lymph node metastasis.(2) The expression of MMP-2and Ki67in esophageal squamous cell carcinoma issignificantly upregulated. The expression of the two protein were related with the degreeof malignancy, TNM staging, clinical stages and the lymph node metastasis. And they haveclosed relationship with invasion, proliferation and metastasis of esophageal carcinoma.(3) There was a positive correlation between the expression of AIB1andMMP-2.They can be used as prediction for invasion and metastasis of esophagealcarcinoma.(4) There was a positive correlation between the expression of AIB1and Ki67.Ki67may be also one of the related protein in AIB1protein non-steroid hormone receptorpathway.
