The collagen which was extracted from pig skin was used as the raw material to prepare collagen hydrolysate.Respectively,investigated its physical and chemical properties, antioxidant activity of different molecular weight fractions in vitro, antioxidant effect in vivo, as well as antioxidant used in lard and pork minced, provided the experimental basis for the further use of pig skin collagen to develop antioxidant peptides.1. Solubility, oil absorption, emulsification, emulsion stability, foaming capacity and foaming stability of the physical properties of pig skin collagen hydrolysate were detected, to provide a certain basis to use it in areas such as food, health products and so on.2. Useing different centrifuge tubes to separate the pig skin collagen hydrolysate, obtained four molecular weight peptides:the molecular weight than lOkDa,5kDa-10kDa,3kDa-5kDa and less than3kDa.The antioxidation of the four molecular weight peptide and pig skin collagen hydrolysate were studied by oxygen free radicals (O2-·) scavenging activity,hydroxyl radical (·OH) scavenging activity,DPPH· radical scavenging activity, reductive ability, inhibition of lipid peroxidation.The results showed that, different molecular weight range of collagen hydrolysate had a certain antioxidant capacity,and antioxidant effect increased with concentration increasing.The hydrolysate showed the highest O2-·scavenging activity, the molecular weight5kDa-10kDa component showed the highest·OH scavenging activity, reductive ability and inhibition of lipid peroxidation, the molecular weight3kDa-5kDa showed the highest DPPH·radical scavenging activity.3. Kunming kinds of mice were randomly divided into5groups,the blank group, the model group,the pig skin collagen hydrolysate low (100mg/kg),medium (200mg/kg) and high (400mg/kg) dose groups.In addition to the blank group,other groups were neck subcutaneous injected with D-galactose. After keeping42days, the activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX) and the content of malondialdehyde (MDA) in serum, liver and brain of the mice were tested. The results showed that, compared with the blank group, the activity of SOD and GSH-PX in the model group were significantly reduced, the content of MDA was significantly increased; Pig skin collagen hydrolysate could increase the activity of SOD and GSH-PX and reduce the content of MDA in serum, liver and brain of the mice, and showed a dose-response relationship.Pigskin collagen hydrolysate has antioxidation effects on aged mice induced by D-galactose.4. Different amounts of pig skin collagen hydrolysate were added to the lard and pork minced to examine its inhibitory effect on oxidation of lard and pork minced.The results show that, the pig skin collagen hydrolysate had a centain antioxidant activity on lard and pork minced, and the antioxidant activity was better with the amount of the pig skin collagen hydrolysate increased, when the amount was2%, it showed the best antioxidant effect, but less than the butyl hydroxy anisd (BHA) of0.02%.
