ObjectiveTo investigate the effects of tuarine on atherosclerosis by observing thechanges in cells with microscope in atherosclerosis models and discuss theanti-inflammatory mechanisms of taurine by testing the diversification of MIFIL-8and TNF-α.Methods32rabbits were randomized divided into normal blank group,model controlgroup,high-dose group and low-dose group. All groups except the normal groupwhich were fed with standard chow diet were given high cholesterol food(perkilo normal food added1%cholesterol,7.5%egg yolk powder,8%pig oil)but thelast two groups added different levels of taurine(4、2g/d). All the animals weresacrificed at12weeks to take the most outstanding aortic atherosclerosisarea.The vassals of the models were observed by light microscope while MIFand IL-8were detected with immuohistochemistry and TNF-αwas tested byenzyme-linked immunosorbent assay.Results①Pathological changes: The aortic of normal blank group was smoothand continuous and had no plaque formation, medial smooth muscle cellsarranged regularly.In aortic arch of model control group,vascular endothelialcell was swelling,denaturation, and irregular thickening as well as contained multi-layer lipid-rich foam cells,the arrangement of smooth muscle cells wasdisordered. Taurine treatment group had clear structure, part of the aortic intimawas thickening, formed witha bit of foam cells and lipid deposition,smoothmuscle cells was still neatly arranged.②Effects of taurine on thickness of aorticintima,mid-membrane,intima/mid-membrane: compared with normal blank gro-up, the intimal thickness, intima/media thickness were significantly increased inthe last three groups(P<0.01);compared with the model control group,the intima/media thickness were significantly lower in the taurine group,(P<0.01). Thedifferences were statistically significant.③MIF and IL-8observtion: blank grouphad rare brown yellow granular (positive staining)substances; there were plentyof brown yellow granular substances in the model control group, part fused intoa sheet shape; high-dose group had very small amout of brown yellow granularsubstaces while the low-dose group had less than it.④Effect of Taurine on theExpression of MIF,IL-8and TNF-α:compared with the normal blank group,the expression of them were significantly increased in the last threegroups(P<0.01);compared with the model control group,the expression weresignificantly lower in the taurine group,(P<0.01).;compared with the high-dosegroup,the expression of MIF,IL-8and TNF-α were increased in the low-dosegroup(P<0.05),The differences were statistically significant.Conclusions1)Expression of MIF,IL-8and TNF-α in atherosclerosis model were high.2)The role of taurine on anti-atherosclerosis of rabbit model maybe dependon decrease the expression of inflammatory factors such as MIF, IL-8andTNF-α,and that’s dose-related.
