| ObjectiveTo prepare the specific egg yolk immunoglobulin against Prevotellaintermedia (Pi) which is an important Pathogen of pregnancy gingivitis andevaluate its activity in vitro and rat.MethodsLaying hens were immunized with formaldehyde killed Pi. IgY was isolatedand purified from egg yolks by water dilution, salt precipitations andultrafiltration. Growth inhibition of P. intermedia–IgY at various concentrationsto Pi was tested in liquid culture medium. SD rats were divided into severalgroups randomly and modeled to gingivitis-causing Prevotella intermedia (Pi).Gingival index(GI)，plaque index(PI)，bleeding on probing(BOP)，white bloodcell count(WBC) and weight of the rats were measured. Tissue slices of gumfrom inferior teeth were prepared to observe local tissue inflammation.Results1.P. intermedia–IgY inhibited the growth of Pi in a dose-dependent manner.P. intermedia–IgY at a concentration of20mg/ml inhibited the growth ofbacteria completely. Its inhibition activity was similar to that of tinidazole(p<0.05).2. Compared with blank control group, the GI, PI, BOP and WBC of rats inother control groups decreased significantly(p<0.05)while weight increasedsignificantly(p<0.05), and the sections showed lighter local inflammation.Compared with positive control group, rats in the treatment groups showed much lower GI, PI, BOP and WBC (p<0.05), much higher weight(p<0.05),and much lighter local inflammation. Treatment by IgY solution group showedbetter efficacy than yolk powder group (p<0.05).Conclusions1. P. intermedia–IgY can inhibit the growth of Pi indicating the potential ofspecific IgY to be an alternative therapy for pregnancy gingivitis caused by Pi.2. Both forms of P. intermedia–IgY are effective on treatment of ratgingivitis caused by Pi. IgY solution is better than yolk powder. |
