Objective: Preparation and purification of specific yolk immunoglobulin(Ig Y)against Prevotella intermedia(P.intermedia,Pi)and evaluation of its therapeutic effect on experimental gingivitis in pregnant ratsMethods: Preparation of specific Ig Y by Two-step ammonium Sulfate precipitation method,Twenty-four female SD rats were randomly divided into 4 groups: group A(blank control group);group B(negative group): modeled+salt saline;group C:(positive control group)modeled + tinidazole solution;group D:(experimental group)modeled + specific Ig Y solution.Use silk thread to ligate the upper and lower first molars.Drinking 10% sucrose water,inoculating bacteria,After 1 week of ligature,2:1at the same time as the normal group,and caged with male rats.The therapeutic effect of specific Ig Y was observed by gingival index(GI),plaque index(PLI),body weight change,RT-q PCR detection of the composition ratio of P.intermedia in plaque before and after treatment,and histopathological sections.Results:Through the treatment of experimental pregnancy gingivitis in rats,it was found that the GI,PLI(P<0.001)and body weight(P<0.05)of the 2.5 mg/ml tinidazole group and the Ig Y treatment group were improved compared with the saline group,and The composition ratio of P.intermedia was significantly lower in the subgingival plaque in the tinidazole group and the Ig Y group(P<0.05),but there was no significant difference before and after treatment in the saline group(P>0.05).Histopathological observation of the gingiva showed that the inflammatory state of the gingiva in the anti-Ig Y group and the tinidazole group was lighter than that in the saline group.Conclusion: Anti-P.intermedia-specific yolk antibody can improve gingival inflammation in rats during pregnancy.
