Most of the plant source of alkaloids with medicinal activity, it is a class earliest study of the most successful and study a large class of natural products.Existing research Scorzonera divaricata with a variety of pharmacological live, few studies on the separation and purification of its chemical composition, the alkaloids in the determination of the level of alkaloids in chemical composition and structure of identification and biological activity has not been reported, the active ingredients of its alkaloids isolation, purification and biological activity of a fundamental role.Scorzonera divaricata aerial parts were investigated in this article. The key content were that determination, purification technology and antioxidant activity of total alkaloid in S.divaricata.This research is aim to discover the isolation and purification of alkaloids of it, and make an primary study on its antioxidate activities, so that to laid a theoretical foundation for the utilization of S.divaricata and drug design. Results are as follows:1. An alkaloid individual composition study method was used to identify the alkaloid chemical compounds in S. divaricata.2. Established a acid-dye colorimetry for the assay of alkaloids in S.divaricata, and the Appropriate conditions as follows:the buffer is pH4.6,the bromocresol green buffer useage is6mL and chloroform useage is10mL, Shaking uniform and then standing, extracted by chloroform,the layer of chloroform after drying, detecting at430nm. Under these conditions, the centent of total alkaloids is0.808mg/g.3. In this experiment, macroreticular resin is used in purification technology of total alkaloids in S. divaricata. From the comparison of five macroreticular resins which are of different polarities in their adsorption and desorption ability, the HP-20macroporous resin is selected. By further optimization, the optimal conditions were follows:the sample pH of8.0, total alkaloids concentration in the solution of0.5mg/mL, the volume of sample is20mL(2g), washing with70%ethanol.After the purifition, thepurity of alkaloids rised from27.6%to64.7%.4. The total alkaloids extraction was separated by silica gel column chromatography, further purified by repeated silica gel and LH-20gel column chromatography, get a white powder compound (3-sitosterol.5. In this experiment detemined the total alkaloids reduce ability and ability to scavenge DPPH·, ABTS+and·OH. The results showed that S.divaricata total alkaloid have a certain restore ability, with the DPPH·, the ABTS+. hydroxyl radical scavenging activity, the reducing ability, free radical scavenging ability with its concentration was linear positive correlation.
