This thesis used Scorzonera divaricata. L as experimental material, studied extraction progress, deproteinization and decolouration technology, antioxidation activity in vitro and antibacterial and antifatigue ability of water-soluble polysaccharides from Scorzonera divaricata. L.1. Orthogonal experiment was used to optimize the technological conditions of the traditional Water circumfluence method to extract Scorzonera divaricata. L polysaccharides, to confirm the best parameter. Results showed:the affect order of different factors was temperature>time>solid-liquid ratio>times, the optimal conditions were time of90min, temperature of90℃, solid-liquid ratio of1:35and times of3.Under the best conditions, the extraction ratio of polysaccharides could reach to6.43%.2. Uniform design was used to optimize the technological parameters of modern microwave-assisted method to extract Scorzonera divaricata. L polysaccharides, to determine the optimum variables. Results showed:the optimal variables were microwave power246w, solid-liquid ratio1:39, extracting time60min, extraction times2. Under the best conditions, the extraction rate of polysaccharides could reach to3.37%.3. Sevage method, trichloroacetic acid method and hydrochloric acid method were used separately to remove the protein of Scorzonera divaricata. L polysaccharides, orthogonal experiment was adopted to optimize the optimal deproteinization method. Results showed:the Sevage method was better than other ways; the optimal technological conditions were oscillation time25min, chloroform:normal butanol3:1, sample:Sevage5:2and adding times6. Under the best conditions, the deproteinization rate could reach to71.26%.4. Activated carbon method, sodium hypochlorite method and hydrogen peroxide method were used respectively to decolour the Scorzonera divaricata. L polysaccharides, uniform design was used to optimize the best decolouration way. Results showed:activated carbon method had better effect than others; the optimal variables were solid-liquid rate0.53:50, water bath time47.36min, temperature37.49℃and pH3.96. Under the optimized conditions, the decolouration ratio could reach to91.88%.5. Spectrophotometry was used to determine the scavenging capacities of Scorzonera divaricata. L polysaccharides on DPPH, hydroxyl and superoxide anion free radicals, and its reducing power and chelating ability. Rancimat method was used to assay its anti-lipid peroxidation ability. Results showed:It had certain remove ability on free radicals, and had obvious dose-effect relationship in the concentration range; It was not suited to use as iron ion chelating agent; It could be as antioxidant added in lard, sunflower seed oil and cotton seed oil after further processing, while not be as anti-lipid peroxidation additive of rapeseed oil.6. Beating holes method was used to study the bacterial activity of Scorzonera divaricata. L polysaccharides on the19common microbial strain. Results showed:It had bad bacterial ability, so it could not be developed as microbial growth inhibitors.7. Mice experiment of swimming with weight was used to explore the antifatigue function of Scorzonera divaricata. L polysaccharides. Results showed:compared with the blank group, the control group, the low-dose group, the middle-dose group and high-dose group added respectively the mice swimming time by2.5%,11.7%,20.4%,35.1%.
