Objective:To determine the effect s o f pseudolaric acid B, PAB on the Proliferation of ratglioma C6cells.Methods:PAB (10,25,50μmol/L) was added into the culturedC6cell s and incubated for24,48and72h. The changes of cell morphology were observed by phase contrastmicroscope. MTT assay was used to detect the proliferationrate of C6cells,Cell apoptosis were analyzed by flowcytometry,Detection pseudolaric acid to the growth of the tumor of nude mice after inoculation of C6glioma cells. Measure the volume, of tumor,after32days killthenude miceand take the weight of the tumor.Results:Various concentrations of PAB inhibited the growth of C6cells in a dose-time-dependent manner.C6cells treated with2.0.4.0.8.0μmol/L PAB for24h showe significantly enhance dapoptosisas compared with the controlcellsPseudolaric acid can inhibit the proliferation of C6cells in vivo.Conclusions:PAB inhibit The growthof glioma C6cell.The inhibitory rate and the apoptotic cell rate were related with the time and medicine concentration.. Pseudolaric acid can inhibit the proliferation of C6cells in vivo.
