| Objective:The aim of this study was to investigate the effect of Pseudolaric acid B on cell proliferation,cell cycle and apoptosis of FaDu cells and its possible mechanism.Methods:The following FaDu cell lines of hypopharyngeal cancer were used as the research objects.Different concentrations(0.000,0.125,0.250,0.500,1.000,2.000,4.000,8.000 pmol/L)of Pseudolaric acid B were used to act on FaDu cells of human hypopharyngeal cancer respectively.CCK-8 method was used to detect the cell proliferation after 24,48,72 hours.SPSS software was used to calculate the IC25,IC50,IC75 values.Different concentrations(0.000,0.324,1.640,8.307 μmol/L)of Pseudolaric acid B were used for 48 hours of treatment,the cell cycle distribution and apoptosis of FaDu cells were detected by flow cytometry,and the expression of apoptosis related proteins Bcl-2,Bcl-xl and Bax were detected by Western blot.The data were analyzed by spss19.0.Results:CCK-8 method showed that Pseudolaric acid B could significantly inhibit the proliferation of human hypopharyngeal cancer FaDu cells.With the increase of drug concentration(0.125,0.250,0.500,1.000,2.000,3.000,4.000,8.000μmol/L)and action time,the survival rate of hypopharyngeal cancer FaDu cells gradually decreased,and the difference was statistically significant(P<0.05),showing a concentration and time-dependent.The IC50 was 1.640 μmol/L.The results of flow cyd tometry showed that the cell cycle arrest ate of 0.324 μmol/L,1.640μmol/L and 8.307 μmol/L Pseudolaric acid B treated for 48h was significantly higher than that of the control group(P<0.05).The results of flow cytometry showed that the apoptotic rate of cells treated with 0.324 μmol/L,1.640 μmol/L and 8.307μmol/L Pseudolaric acid B for 48 hours was(24.47±1.08)%,(44.32±2.47)%,(72.55±0.93)%,respectively,which was significantly higher than that of the control group(2.73±1.37)%(P<0.001).The results of Western blot showed that the expression of Bax in FaDu cells increased with the increase of the concentration of Pseudolaric acid B(0.324 μmol/L,1.640μmol/L,8.307 μmol/L),but the expression of Bcl-2 and Bcl-xl decreased,the difference was statistically significant(P<0.001).Conclusion:Under certain conditions,Pseudolaric acid B can inhibit the proliferation of FaDu cells,block the cell cycle of tumor cells in the G0/G1 phase,and promote cell apoptosis.The mechanism may be related to the up-regulation of Bax expression and down-regulation of Bcl-2 and Bcl-xl expression. |
