Objectives:Dioctylphthalate(DOP) was largely used as plasticizer and additive in plastic,pesticide, rubber and cosmetic productions, but it is likely for DOP to release intoenvironment and cause estrogen-like effects, thus to be considered as a pollution.Chloroform (CF) is the byproduct of chlorination disinfection, which is also harmfulto human body, there was few reports on their combined effect. This study observedmice under DOP and chloroform combined exposure, recorded their body weight,organ coefficient, lipid peroxidation, enzyme activities, calcium ion levels andmitochondrial membrane potential in their brain cells, so as to study the combinedeffects of the two agents on brains.Method:This study was designed corresponding the analysis of combined effect,2×2factorialanalysis. The two factors were DOP and chloroform, exposure group andnon-exposure group were crossed, resulting the grouping of control group, DOPexposure group, chloroform exposure group, combined exposure group.Mice were exposed via gavage through gastrointestinal tract, mice wererandomly grouped as control group (corn oil), DOP exposure group(1/10LD50,540mg/kg), CF exposure group(1/10LD50,112mg/kg) and DOP(1/10LD50,540mg/kg)+CF(1/10LD50,112mg/kg) exposure group. Mice were exposed every day for15days, after when they were euthanized, their brain organ coefficient, neurotransmitter,lipid peroxidation index, enzyme activity, calcium ion level and mitochondrialmembrane potential were examined.Results:1、Effect of Dioctylphthalate combined with Chloroform exposure on the body weight ofmice.Gavage infected after15days, compared with the control, the body weight offemale mice in DOP group increased (P<0.05), combined exposure group existedinteraction effect (P<0.05); the body weight of male mice in chloroform groupreduced (P<0.05). 2、Effect of Dioctylphthalate combined with Chloroform exposure on lipid peroxidation ofmice brain.Compared with the control, the activity of CAT in DOP group increased in thefemale mice (P<0.05), combined exposure group e of mice xisted interaction effect (P<0.05);the content of MDA in chloroform group reduced in the male mice (P<0.05), combinedexposure group existed interaction effect (P<0.05).3、Effect of Dioctylphthalate combined with Chloroform exposure on neurotransmitter of micebrain.Compared with the control, the content of5-HT、NE in DOP and Chloroformgroup reduced in the female mice (P<0.05), combined exposure group existedinteraction effect (P<0.05); the content of DA in chloroform group reduced (P<0.05),combined exposure group existed interaction effect (P<0.05). The content of5-HTin DOP group increased in the male mice (P<0.05), chloroform group reduced(P<0.05); the content of NE in combined exposure group existed interaction effect(P<0.05); the content of DA in DOP and Chloroform group increased in the malemice (P<0.05), combined exposure group existed interaction effect (P<0.05).Theeffect of DOP combined with Chloroform exposure on neurotransmitter of mice brain, inaddition to the5-HT in control group there were gender differences(P<0.05).4、The effect of DOP combined with Chloroform exposure on neurotransmitter onNa+-K+-ATPase and Ca2+-Mg2+-ATPase of mice brain.Compared with the control, the both activity of female mice in DOP andChloroform group increased (P<0.05), Ca2+-Mg2+-ATPase of combined exposuregroup existed interaction effect (P<0.05); the both activity of male mice in Chloroformgroup increased (P<0.05), and combined exposure group both existed interaction effect(P<0.05).5、The effect of DOP combined with Chloroform exposure on neurotransmitter on LDH、SDH、NOS、NO of mice brain.Compared with the control, the activity of LDH、SDH in Chloroform groupincreased in the female mice (P<0.05), combined exposure group both existedinteraction effect (P<0.05); the content of NO in DOP group reduced (P<0.05),combined exposure group existed interaction effect (P<0.05). The activity of LDH、SDH in DOP group reduced in the female mice (P<0.05), combined exposure group both existed interaction effect in DOP and Chloroform group (P<0.05); the activity ofNOS in Chloroform group increased (P<0.05), the content of NO in DOP groupreduced (P<0.05).6、The effect of DOP combined with Chloroform exposure on the calcium ion level andmitochondrial membrane potential of mice brain.Compared with the control, the calcium ion level of female mice in DOP andChloroform group reduced (P<0.05), mitochondrial membrane potential increased (P<0.05),the calcium ion level in combined exposure group existed interaction effect (P<0.05); themitochondrial membrane potential of male mice in Chloroform group increased (P<0.05),combined exposure group existed interaction effect (P<0.05).Conclusion:1、At the same time contact DOP and chloroform could cause the activity of CATincreased in the female mice, the activity of CAT reduced and the content of MDAincreased in the male mice.2、The content of5-HT、NE、DA increased in the male mice, the female mice opposite,the role of neurotransmitter in a certain gender differences between male and female.3、The activity or content of LDH、SDH、NOS、NO、Ca2+-Mg2+-ATPase increasedin the female mice brain, the activity of LDH、SDH reduced, Na+-K+-ATPase andCa2+-Mg2+-ATPase of female mice increased.4、The calcium ion level reduced, the mitochondrial membrane potential of male miceincreased.5、DOP combined with Chloroform exposure can produce interaction effect on mice brain.
